Immunomodulatory effects and antimicrobial activity of heterofucans from Sargassum filipendula

Parasitic diseases are a human health problem mainly in low-income areas. The drugs available for the treatment of these diseases are far from satisfactory due to high costs, toxicity, and drug resistance. Sulfated polysaccharides are a complex group of bioactive polymers and can be obtained from seaweeds. The heterofucans from Sargassum filipendula (SF) present strong antiproliferative and antioxidant activities. However, their immunomodulatory and antimicrobial capacity have not been evaluated until now. In this study, five sulfated fucose-rich fractions were isolated (named SF0.5V, SF0.7V, SF1.0V, SF1.5V, and SF2.0V). The chemical composition showed slight differences among polysaccharides and, consequently, biological activity of these polymers. Three fractions (SF0.5V, SF0.7V, and SF1.0V) showed a strong immunomodulatory activity enhancing the release of nitric oxide (NO) by murine macrophages (RAW 264.7), though only SF0.5V was able to induce interleukin-6 (IL-6) and TNF-alpha release from RAW cells. The sugar to sulfate ratio was not correlated with these activities. Meanwhile, the contents of xylose (P = 0.98 for NO; P = 0.98 for IL-6; P = 0.96 for TNF-alpha) and glucuronic acid (P = 0.91 for NO; P = 0.9190 for IL-6; P = 0.79 for TNF-alpha) were strongly positively correlated. SF0.7V and SF1.0V inhibited biofilm formation by Klebsiella pneumoniae (4.2 and 6.8%, respectively), whereas SF0.5V showed inhibitory effect (similar to 50%) on biofilm formation by Staphylococcus epidermidis. Furthermore, SF0.7V and SF1.0V showed high inhibition capacity on the survival of the protozoan Trichomonas vaginalis. The sugar to sulfate ratio was positively correlated (P = 0.60) with this activity. The results demonstrate the spectrum of action of these sulfated polysaccharides obtained from SF and show their potential as immunomodulatory and microbicidal agents.

Antioxidant Activity of Fucoidan Modified with Gallic Acid Using the Redox Method

Summary: Antioxidant compounds reduce intracellular reactive oxygen species (ROS) and protect osteoblasts from ROS-induced damage. This study demonstrates that modifying a 21 kDa fucoidan with gallic acid can enhance its antioxidant capacity and protect pre-osteoblast-like cells against oxidative stress. The modified FucA-GA showed significantly higher antioxidant activity compared to other compounds, effectively reducing ROS levels and caspase activation. It also demonstrated promising protective effects against oxidative damage in zebra fish embryos. Overall, FucA-GA could have potential therapeutic applications for bone fragility by counteracting oxidative stress.

Comparison of in vitro and in vivo antioxidant activities of commercial fucoidans from Macrocystis pyrifera, Undaria pinnatifida, and Fucus vesiculosus

Summary: This study compared the antioxidant activities of three fucoidans from different seaweeds, and found that fucoidan from Macrocystis pyrifera exhibited the highest antioxidant activity. The fucoidans had low electron donation capacity and metal chelating ability, but showed good scavenging activity against hydroxyl radicals. In cell and zebrafish embryo experiments, all fucoidans protected cells from oxidative damage.

Essential Oils and Terpenic Compounds as Potential Hits for Drugs against Amitochondriate Protists

Summary: This article summarizes the human anaerobic or microaerophilic protists Giardia duodenalis, Entamoeba histolytica, and Trichomonas vaginalis which are classified as amitochondriate parasites. These microorganisms have adapted to survive in hostile microenvironments and pose an increasing threat to public health in developing countries. Current therapeutic drugs to manage these infections are scarce and often have several side effects. This review summarizes essential oils and terpenic compounds with activity against these amitochondriate parasites and explores possible mechanisms of action. It aims to contribute to future research perspectives on using natural products as potential alternatives for treating amitochondriate protists.

Chondroitin Sulfate from Oreochromis niloticus Waste Reduces Leukocyte Influx in an Acute Peritonitis Model

Summary: Oreochromis niloticus (tilapia) is a commonly cultivated fish species, and its farming generates organic waste. In this study, tilapia viscera were processed to obtain chondroitin sulfate (CST), a biomolecule that showed potential as an anti-inflammatory drug. The CST obtained from tilapia waste significantly reduced leukocyte migration in an acute peritonitis model, indicating its pharmacological activity.

Biological and pharmacological aspects of tannins and potential biotechnological applications

Summary: This article explores the characteristics of tannins and their role in regulating ROS and maintaining homeostasis. Tannins have anti-inflammatory, anti-aging, and anti-proliferative properties due to their ability to bind with reactive oxygen species. They can reduce the risk of cancer and neurodegenerative diseases and also maintain a healthy gut microbiota. Tannins have industrial applications in leather durability, metal corrosion resistance, 3D printing, and bio-based foam manufacturing. This review highlights the biological and pharmacological relevance of tannins and their potential applications.

Summary: The objective of this study was to generate blends containing SPGb and ChrPic that exhibit enhanced antioxidant activity compared to SPGb or ChrPic alone. B5 was identified as the best antioxidant blend, which showed no cytotoxicity and genotoxicity. These findings suggest that B5 could be further tested in vivo to confirm its antioxidant action and potentially used as a nutraceutical agent.

Summary: Medicinal plants, such as Talisia esculenta, have been found to contain antioxidant biomolecules that can be used in the treatment and prevention of diseases. This study investigated the antioxidant potential of T. esculenta extracts from leaves and fruit peels using various assays. The extracts exhibited antioxidant activity, protecting against oxidative stress and containing phenolic compounds. These findings suggest that T. esculenta extracts could be used as coadjutants in the development of natural nutraceutical products for the treatment and prevention of diseases related to oxidative stress.

Comparative analysis of the biocompatibility of endothelial cells on surfaces treated by thermal plasma and cold atmospheric plasma

Summary: This study evaluated the effects of cold atmospheric plasma (CAP) and thermal plasma methods on the biocompatibility of titanium surfaces. The results showed that CAP treatment improved the hydrophilicity and reduced the roughness of the titanium surface, enhancing the biocompatibility of endothelial cells. This technique has the potential for biomedical applications due to its low cost and ease of use.
