Summary: In this study, a novel multicolor staining protocol was introduced for the visualization of multiple tissue components in bone samples. The staining method, which involved optimized reagents and procedures, allowed the simultaneous identification of high-mineralized bone, low-mineralized fracture callus, cartilage, and skeletal muscle fibers. The efficacy of the staining method was further validated through immunohistochemical staining and micro-CT analysis. The results demonstrated that the multicolor staining provided quality outcomes with fewer reagents and simplified procedures, making it a valuable tool for future preclinical research involving decalcified paraffin-embedded samples.

Summary: This study reveals for the first time that myofibroblast expression levels are higher in the dorsal layer of the ligamentum flavum (LF) than in the dural layer. It confirms that hypertrophy of the LF in lumbar spinal canal stenosis is associated with increased expression of myofibroblasts in the dorsal layer. Hypoxia could be a cause of myofibroblast expression leading to fibrosis and ultimately to hypertrophy of the LF.

Summary: In this study, X-ray diffraction tensor tomography was used to non-destructively image the orientation of HA in a piglet's medial femoral condyle, revealing local variations and predominantly front-directed orientation. The ability to retrieve 3D orientation maps of bone-cartilage structures is expected to enhance understanding of the physiological properties of bones.

Summary: This study aimed to evaluate the detection and tissue distribution of Treponema pallidum in skin lesions of syphilis. Immunohistochemistry and Warthin-Starry silver staining were used for diagnostic accuracy in skin samples from patients with syphilis and other diseases. The results showed that immunohistochemistry had a sensitivity of 60% and a specificity of 100% in detecting spirochetes in skin samples from patients with syphilis.
