Journey of cystatins from being mere thiol protease inhibitors to at heart of many pathological conditions

Cystatins are thiol proteinase inhibitors (TPI), present ubiquitously in animals, plants and microorganisms. These are not merely inhibitors rather they are at heart of many pathological conditions ranging from diabetes to renal failure. These are essential for maintenance of protein balance of the cell; once this balance gets disturbed, it may lead to cell death. Thus, cystatins cannot be merely regarded as TPI's as these have been found to play a pivotal role in tumorigenesis and neurodegenerative diseases. Many studies have reported the variation in cystatin level in incidences of different types of cancer; highlighting an important role played by these inhibitors in cancer development and progression. Cystatin C is increasingly replacing creatinine as a biomarker of glomerular filtration rate (GFR) thereby highlighting the importance of this important inhibitor. Some recent studies have also reported the interaction pattern of various anti-cancer drugs with cystatins in a bid to find how these drugs affect this important inhibitors and whether these drugs have any side effect on cystatins. Thus, in this growing disease era it can be said that cystatins are no more just inhibitors blocking the activity of thiol proteases rather they play a pivotal role in variety of pathological conditions. Summary: Proto-oncogene tyrosine-protein kinase ROS (ROS1) is a member of a receptor family that affects epithelial cell differentiation and is highly expressed in various tumors. This study identified two potential inhibitors of ROS1 through virtual screening of phytochemicals. The findings highlight the role of ROS1 in multiple physiological processes and suggest the potential use of phytochemicals for therapeutic development targeting ROS1-associated non-small cell lung cancer.

Investigating the Effect of Cigarette Smoking on Serum Uric Acid Levels in Multiple Sclerosis Patients: A Cross Sectional Study

Summary: This study quantitatively examines the effect of cigarette smoking on serum uric acid levels in people with multiple sclerosis (pwMS) before and after smoking cessation. It also investigates the possible correlation between uric acid levels and disability progression and disease severity. The results show that smokers with pwMS have significantly lower serum uric acid levels compared to non-smokers, which can be restored after smoking cessation. However, there is no correlation between serum uric acid levels and disability or disease severity in current smokers with pwMS.

Linagliptin and Empagliflozin Inhibit Microtubule Affinity Regulatory Kinase 4: Repurposing Anti-Diabetic Drugs in Neurodegenerative Disorders Using In Silico and In Vitro Approaches

Summary: Type 2 diabetes mellitus (T2DM) and Alzheimer's disease (AD) have a common pathophysiology, and discovering the action mechanism of anti-diabetic drugs in AD and related pathologies is in high demand. In this study, we used structure-based virtual screening to identify potential MARK4 inhibitors among FDA-approved anti-diabetic drugs. We found that linagliptin and empagliflozin have strong affinity and specificity towards the binding pocket of MARK4 and showed significant inhibition of MARK4 kinase activity.

In vitro and In vivo study targeting the development of Unani Antidermatophytic Cream: Implication of Herbal Formulations in Treatment of Dermatophytosis

Summary: Panwad, Sarshaf, and Kunjad have been traditionally used for treating ringworm infection and have shown promising results. A cream containing extracts of these plants was developed and evaluated for better efficacy. The final batches of the cream demonstrated significant antifungal activity in vitro and in vivo. It can be concluded that this cream can be an alternative topical agent for treating dermatophytosis.

Investigating molecular interactions between human transferrin and resveratrol through a unified experimental and computational approach: Role of natural compounds in Alzheimer's disease therapeutics

Summary: Disruptions to iron metabolism and iron homeostasis are important factors in the development of Alzheimer's disease (AD). Human transferrin plays a crucial role in maintaining iron balance in the body. Plant-derived compounds and dietary constituents, particularly polyphenols found in fruits, vegetables, teas, coffee, and herbs, show promise in preventing AD. Resveratrol, a natural polyphenol found in grapes, berries, peanuts, and red wine, exhibits neuroprotective effects that may help prevent or treat AD through various mechanisms. Molecular docking and molecular dynamic simulations were used to study the interaction between transferrin and resveratrol, confirming their binding. These findings provide insights into the potential therapeutic applications of natural compounds, particularly in targeting iron homeostasis in AD.

Phytoconstituents as potential therapeutic agents against COVID-19: a computational study on inhibition of SARS-CoV-2 main protease

Summary: The COVID-19 pandemic caused by SARS-CoV-2 is a global health crisis, and effective treatments are urgently needed. In this study, through computational screening and validation, we identified two phytoconstituents, Khelmarin B and Neogitogenin, as potential inhibitors for the main protease of SARS-CoV-2, which is essential for viral replication. These compounds show promising binding affinities and specificity for the target, suggesting their potential as COVID-19 therapeutics. Further experimental studies are required to confirm their efficacy and safety.

Structure-guided design and development of vanillin-triazole conjugates as potential MARK4 inhibitors targeting hepatocellular carcinoma

Summary: This study synthesized vanillin-triazole derivatives and found that compounds 5a, 5g, 5h, and 5j showed significant inhibitory activity on MARK4 enzyme. Molecular docking and MD simulation studies confirmed their strong binding with MARK4 and their ability to suppress cell proliferation, migration, and induce apoptosis in hepatocellular carcinoma cells. These findings suggest that vanillin-triazole compounds, particularly compound 5g, could be potential candidates for developing MARK4-based anticancer therapies.

Therefore, there is increasing interest in using anti-inflammatory compounds and antioxidants to target neuroinflammation. This study investigates the binding and dynamics of the human transferrin (HTF)-apigenin complex, and suggests that apigenin has a neuroprotective effect by binding to HTF and maintaining a stable conformation.

Identification of mitogen-activated protein kinase 7 inhibitors from natural products: Combined virtual screening and dynamic simulation studies

Summary: Mitogen-activated protein kinase 7 (MAPK7) plays a vital role in various cellular processes and its elevated expression has been connected to aggressive tumors. This study used virtual screening to identify potential MAPK7 inhibitors from natural products and identified two compounds, ZINC12296700 and ZINC02123081, with strong binding affinity and drug-like properties. Molecular dynamics simulations confirmed the stability of the docked complexes, and molecular mechanics calculations indicated considerable binding affinities. These compounds hold promise as lead candidates for developing novel MAPK7 inhibitors.

Effects of aging and diabetes on the deformation mechanisms and molecular structural characteristics of collagen fibrils under daily activity

Summary: Crosslinking affects the mechanical behavior and tissue metabolism of collagen-based tissues. Aging and diabetes alter the crosslinking type and density, thus changing tissue properties. High-connectivity enzymatic crosslinking and advanced-glycation end-products result in uniform deformation under daily activity, while low-connectivity enzymatic crosslinking does not. High-connectivity model displays more sliding, while AGEs induce instability in structures near the binding sites.

An eco-friendly composite hydrogel based on covalently crosslinked cellulose/poly (glycerol citrate) for thallium (І) removal from aqueous solutions

Summary: In this study, a reusable biobased composite hydrogel was successfully synthesized and used for the removal of Tl(I) from aqueous solutions. The hydrogel exhibited high adsorption capacity and thermal stability, and the adsorption process was found to be thermodynamically spontaneous. The results demonstrated that the hydrogel can be reused multiple times without significant loss of its adsorption capacity.

The characteristics and optimal conditions of the adsorbent were assessed and analyzed.

Furthermore, the use of chitosan and dextran sulfate as wall materials in the multi-layer nanoemulsions led to even higher cellular uptake, in vitro bioaccessibility, and antioxidant activity of alpha-tocopherol.

Stabilization of oil-in-water high internal phase emulsions with octenyl succinic acid starch and beeswax oleogel
