Multi-disciplinary summit on genetics services for women with gynecologic cancers: A Society of Gynecologic Oncology White Paper

Objective. To assess current practice, advise minimum standards, and identify educational gaps relevant to genetic screening, counseling, and testing of women affected by gynecologic cancers. Methods. The Society of Gynecologic Oncology (SGO) organized a multidisciplinary summit that included representatives from the American College of Obstetricians and Gynecologists (ACOG), the American Society Clinical Oncology (ASCO), the National Society of Genetic Counselors (NSGC), and patient advocacy groups, BrightPink and Facing our Risk of Cancer Empowered (FORCE). Three subject areas were discussed: care delivery models for genetic testing, barriers to genetic testing, and educational opportunities for providers of genetic testing. Results. The group endorsed current SGO, National Comprehensive Cancer Network (NCCN), and NSGC genetic testing guidelines for women affected with ovarian, tubal, peritoneal cancers, or DNA mismatch repair deficient endometrial cancer. Three main areas of unmet need were identified: timely and universal genetic testing for women with ovarian, fallopian tube, and peritoneal cancers; education regarding minimum standards for genetic counseling and testing; and barriers to implementation of testing of both affected individuals as well as cascade testing of family members. Consensus building among all stakeholders resulted in an action plan to address gaps in education of gynecologic oncology providers and delivery of cancer genetics care. Summary: PARP inhibitors are effective in germline BRCA1/2-mutated metastatic breast cancer. This study demonstrates that adding veliparib to cisplatin improves progression-free survival in patients with BRCA-like metastatic triple-negative breast cancer.

Executioner caspases restrict mitochondrial RNA-driven Type I IFN induction during chemotherapy-induced apoptosis

Summary: During apoptosis, the release of mitochondrial RNA into the cytosol can be prevented by casp3/7, which inhibits inflammatory signaling. Inhibition of casp3/7 promotes a cytosolic mtRNA-driven Type I interferon response, increasing the immunogenicity of chemotherapy-induced apoptosis.

Fracture strength of screw-retained zirconia crowns assembled on zirconia and titanium implants

Summary: This study evaluated the fracture strength of zirconia crowns connected to zirconia and titanium implants after simulating 5 years of clinical use. The results showed that the zirconia crowns connected to titanium-zirconia and titanium implants had significantly higher fracture strength than those connected to pure zirconia or lubricant zirconia implants. Therefore, zirconia crowns can withstand the common occlusal forces in the anterior and premolar regions.

Fanconi anemia-associated chromosomal radial formation is dependent on POLθ-mediated alternative end joining

Summary: Activation of the Fanconi anemia (FA) pathway is crucial for preventing chromosome translocations referred to as "radials" after treatment with mitomycin C (MMC). Radials induced by MMC are dependent on DNA polymerase theta (POLq)-mediated alternative end joining (A-EJ) in FA-deficient cells, specifically involving POL theta and DNA ligase III. Blocking POLq in FA-deficient cells eliminates radials and leads to the accumulation of breaks at common fragile sites. These findings provide mechanistic insights into the formation of radials and offer potential therapeutic strategies for FA pathway-deficient cancers using POLq inhibitors.

Summary: Mathematical modeling of early CA-125 kinetics can be used as an indicator of rucaparib efficacy in recurrent ovarian carcinoma patients. The KELIM-PARP score is associated with treatment response and progression-free survival. Integration of spatial and single-cell data across modalities with weakly linked features

Summary: This article introduces a cross-modal data integration method called MaxFuse, which utilizes iterative coembedding, data smoothing, and cell matching to achieve high-quality integration even with weakly linked features. The method is modality-agnostic and demonstrates high robustness and accuracy in the weak linkage scenario, achieving a relative improvement of 20% to 70% over existing methods on benchmarking datasets.

Quantifying common and distinct information in single-cell multimodal data with Tilted Canonical Correlation Analysis

Summary: Multimodal single-cell technologies enable the analysis of multiple modalities for each cell simultaneously, providing a more comprehensive characterization of cell populations. Our developed method, Tilted Canonical Correlation Analysis (Tilted-CCA), effectively separates and quantifies shared information among cells and modality-specific information, allowing meaningful visualization and quantification of cross-modal information.
