Background and aims: Hydroxytyrosol (HT) is the primary phenolic compound of olives, virgin olive oil, and their byproducts. Proteomic analysis of metabolically active tissues helps elucidating novel mechanisms of action and potential targets in cardiometabolic disease. Thus, we aimed at determining the impact of long-term HT supplementation on the proteome of adipose and liver tissue, in mice. Methods: C57BL/6J mice received either a control diet or a diet supplemented with nutritionally relevant doses of HT for eight weeks. Results: HT supplementation differentially affects the adipose and liver tissues proteome, as evaluated by super-SILAC. Some oxidative stress-related proteins were modulated in both tissues, such as the multifunctional protein peroxiredoxin 1, which was consistently repressed by HT supplementation. In some cases tissue-dependent modulation was observed, as in the case of FASN. Conclusions: This study provides interesting information on the connection between changes seen at tissue proteome level and the metabolic effects of HT. The use of this pertinent proteomics quantification approach may prove quite useful for uncovering novel potential pharmaco-nutritional targets of HT supplementation. (C) 2017 Elsevier Ltd. The emergent role of mitochondrial RNA modifications in metabolic alterations

Summary: Mitochondrial epitranscriptomics refers to modifications occurring in various types of RNA within mitochondria. These modifications play a critical role in accurate protein synthesis and have implications in mitochondrial-related diseases. Research has focused on mutations in mitochondrial and nuclear DNA genes encoding enzymes involved in these modifications. This article reviews current evidence on mitochondrial RNA modifications and their role in metabolic disorders, and explores the potential of using them as targets for prevention and early detection.

Hydroxytyrosol decreases EDNRA expression through epigenetic modification in colorectal cancer cells

Summary: The Mediterranean diet is associated with lower incidence of cardio-vascular and cerebrovascular diseases as well as cancer. Extra virgin olive oil, with its phenolic components such as hydroxytyrosol, has been attributed with many healthful effects. Recent studies show that extra virgin olive oil and hydroxytyrosol modulate epigenetic mechanisms, specifically DNA methylation, in human tissues. In this study, colorectal cancer cells treated with hydroxytyrosol demonstrated an increase in global DNA methylation, suggesting a potential gene target for this compound.

Dietary Fatty Acids and Inflammation: Focus on the n-6 Series

Summary: Among polyunsaturated fatty acids, the n-3 series (ALA, EPA, and DHA) have been extensively studied for their cardiovascular health benefits, while recent research is focusing on n-6 PUFAs, especially linoleic acid (LA). Although n-6 PUFAs are criticized for being pro-inflammatory, growing evidence suggests their positive effects on cardiovascular health. This review discusses the role of n-6 PUFAs in inflammation, human health, and prognosis, concluding that adequate intakes of n-6 fatty acids are associated with better cardiovascular health and child development.

Integrated transcriptomics and metabolomics analysis reveals the biomolecular mechanisms associated to the antitumoral potential of a novel silver-based core@shell nanosystem

Summary: A combination of transcriptomics and metabolomics was used to study the mechanisms behind the antitumor action of a nanosystem called Ag@MSNs-Tf. Transcriptomics analysis was done using gene microarrays and RT-qPCR, while metabolomics analysis was done using high-resolution mass spectrometry. This multiomics approach revealed the impact of the nanosystem on various molecular pathways, including glycolysis, the pentose phosphate pathway, oxidative phosphorylation, and fatty acid synthesis.

Summary: HT supplementation in humanized mice resulted in systemic dyslipidemia, impaired glucose metabolism, increased body weight, and accumulation of triacylglycerols in different tissues. The expression of genes related to lipid metabolism, such as Pltp or Lpl, was also modulated by HT. Further studies are needed to fully understand the potential effects of HT consumption.
