Serum stearic acid/palmitic acid ratio as a potential predictor of diabetes remission after Roux-en-Y gastric bypass in obesity

Endogenous fatty acid metabolism that results in elongation and desaturation lipid products is thought to play a role in the development of type 2 diabetes mellitus (T2DM). In this study, we evaluated the potential of estimated elongase and desaturase activities for use as predictive markers for T2DM remission after Roux-en-Y gastric bypass (RYGB). The results of a targeted metabolomics approach from 2 independent studies were used to calculate 24 serum FA concentration ratios (product/precursor). Gene expression data from an open public data set was also analyzed. In a longitudinal study of 38 obese diabetic patients with RYGB, we found higher baseline stearic acid/palmitic acid(S/P) ratio. This ratio reflects anelovl6-encoded elongase enzyme activity that has been found to be associated with greater possibility for diabetes remission after RYGB [ odds ratio, 2.16 (95% CI 1.10-4.26)], after adjustment for age, gender, body mass index, diabetes duration, glycosylated hemoglobin A1c, and fasting C-peptide. Our results were validated by examination of postsurgical elovl6 gene expression in morbidly obese patients. The association of S/P with the metabolic status of obese individuals was further validated in a crosssectional cohort of 381 participants. Summary: In this study, a highly sensitive lactate sensor called FiLa was developed, which allows for monitoring subtle lactate fluctuations in living cells and animals. It was found that lactate is highly enriched in mammalian mitochondria and functions as a key hub sensing various metabolic activities. The FiLa sensors also enable direct imaging of elevated lactate levels in diabetic mice and provide a simple, rapid, and sensitive lactate assay for clinical screening. Overall, the FiLa sensors are powerful and broadly applicable tools for studying lactate metabolism in health and disease.

Summary: Sepsis, a global public health challenge, lacks effective treatment or intervention. This study reveals that hyodeoxycholic acid (HDCA) has excellent anti-inflammatory properties in sepsis. HDCA administration significantly decreases systemic inflammatory responses and improves the survival of septic mice. The mechanism involves the competitive blockade of lipopolysaccharide binding to TLR4 and MD-2 receptor complex by HDCA. These findings highlight the potential value of HDCA as a therapeutic molecule for sepsis.

Aqueous cinnamon extract ameliorates bowel dysfunction and enteric 5-HT synthesis in IBS rats

Summary: This study evaluated the effect of cinnamon extract on bowel symptoms in rat models of irritable bowel syndrome with diarrhea (IBS-D). Furthermore, cinnamon extract directly inhibited the expression of Tph1 and controlled serotonin synthesis, providing a scientific viewpoint for the traditional use of cinnamon in treating IBS.

Fecal microbiota transplantation confirmed the link between microbial BA metabolism and intestinal inflammation. The therapeutic potential of ursodeoxycholic acid (UDCA) was evaluated and it was found to alleviate intestinal inflammation through M2 macrophage polarization and suppression of inflammatory cytokines.

ZFP750 affects the cutaneous barrier through regulating lipid metabolism

Summary: An essential function of the epidermis is to provide a physical barrier that prevents water loss. Alteration of ceramides, cholesterol, and very long chain fatty acids, mediators of this barrier function, causes human pathologies. A study shows that genetic deletion of ZFP750 in mice leads to the loss of epidermal barrier function due to a reduction in ceramides. ZFP750 directly and/or indirectly regulates the expression of enzymes involved in ceramide biosynthesis, contributing to our understanding of skin disease pathogenesis.

COVID-19 symptoms and compliance: The mediating role of fundamental social motives

Summary: This study investigates the social motives underlying compliance among individuals with infectious diseases. The results show that individuals with COVID-19 symptoms have lower levels of compliance, which is attributed to a decrease in disease avoidance and familial motives. The study also suggests that exclusion concern weakens the effects of COVID-19 symptoms on compliance. Therefore, enhancing disease avoidance and familial motives may serve as effective intervention strategies to promote compliance among potentially infected individuals.

A method for quantifying hepatic and intestinal ceramides on mice by UPLC-MS/MS
