Purpose MicroRNAs (miRNAs) are short, non-coding RNAs involved in almost all cellular processes. Epigallocatechin-3-O-gallate (EGCG) is a green tea polyphenol and is known to exert anti-arthritic effects by inhibiting genes associated with osteoarthritis (OA). This study was undertaken to investigate the global effect of EGCG on interleukin-1 beta (IL-1 beta)-induced expression of miRNAs in human chondrocytes. Methods Human chondrocytes were derived from OA cartilage and then treated with EGCG and IL-1 beta. Human miRNA microarray technology was used to determine the expression profile of 1347 miRNAs. Microarray results were verified by taqman assays and transfection of chondrocytes with miRNA inhibitors. Results Out of 1347 miRNAs, EGCG up-regulated expression of 19 miRNAs and down-regulated expression of 17 miRNAs, whereas expression of 1311 miRNAs remains unchanged in IL-1 beta-stimulated human OA chondrocytes. Bioinformatics approach showed that 3`UTR of ADAMTS5 mRNA contains the 'seed-matched-sequence' for hsa-miR-140-3p. IL-1 beta-induced expression of ADAMTS5 correlated with down-regulation of hsa-miR-140-3p. Importantly, EGCG inhibited IL-1 beta-induced ADAMTS5 expression and up-regulated the expression of hsa-miR-140-3p. This EGCG-induced co-regulation between ADAMTS5 and hsa-miR-140-3p becomes reversed in OA chondrocytes transfected with anti-miR-140-3p. Conclusions This study provides an important insight into the molecular basis of the reported anti-arthritic effects of EGCG. Our data indicate that the potential of EGCG in OA chondrocytes may be related to its ability to globally inhibit inflammatory response via modulation of miRNAs expressions.

Summary: This study found that honey total polyphenolic fraction (HTPF) has the potential to suppress inflammation in pancreatic islets of Langerhans through the regulation of miRNAs. The researchers also discovered that HTPF inhibits glucose-induced inflammation. These findings provide new insights for using HTPF in the treatment of glucose-induced pancreatic inflammation.

Summary: This study demonstrated that several gene polymorphisms, including ERCC2 rs13181, XRCC1 rs25487, and XRCC3 rs861539, are associated with susceptibility to lung cancer in the Saudi population. The study analyzed 134 lung cancer patients and 270 controls and found significant associations between these gene polymorphisms and squamous cell carcinoma, small cell lung cancer, and adenocarcinoma. Additionally, combined recessive genotypes were also found to be associated with lung cancer susceptibility.

Gene expression of glutathione S-transferase alpha, glutathione S-transferase rho, glutathione peroxidase, uncoupling protein 2, cytochrome P450 1A, heat shock protein 70 in liver of Oreochromis niloticus upon exposure of microcystin-LR, microcystin-RR and toxic cyanobacteria crude

Summary: This study investigated the gene expression variation in the liver of tilapia fish under the effect of MC-LR or MC-RR toxins. The results showed increased expression of GSTA mRNA, variable alterations in GPx, GSTR, UCP2, and CYP1A mRNA, and gradually elevated expression of HSP70 mRNA after MCs exposure. These findings indicate that tilapia liver expresses phase II detoxification enzymes and increased UCP2 and HSP70 expression may protect against oxidative stress and prevent apoptosis.

Aisha Farhana, Abdullah Alsrhani, Nazia Nazam, Muhammad Ikram Ullah, Yusuf Saleem Khan, Zafar Rasheed

Summary: Breast cancer has a high prevalence in females worldwide, and its treatment poses persistent challenges. This study explores the therapeutic potential of gold nanoparticles (AuNPs) in the regulation of microRNA and the protein MMP-9. The results show that AuNPs inhibit MMP-9 expression through the upregulation of microRNA-204-5p in breast cancer cells. Additionally, AuNPs also inhibit NF-?B activation. These findings demonstrate the novel therapeutic role of AuNPs in breast cancer.
