Summary: The study demonstrated that atomoxetine, a clinically approved noradrenaline reuptake inhibitor, improved multiple Alzheimer's disease biomarkers in individuals with mild cognitive impairment. Treatment with atomoxetine significantly reduced CSF levels of Tau and pTau, normalized CSF protein biomarker panels linked to synaptic function, brain metabolism, and glial immunity, and increased brain activity and metabolism in key temporal lobe circuits.

Summary: This study investigates the relationship between the brainstem nucleus locus coeruleus (LC) and functional disturbances in mild cognitive impairment (MCI) patients. It found that functional connectivity (fc) between LC and anterior cingulate cortex and left anterior insula was elevated in MCI patients, while the fc between LC and right temporoparietal junction and posterior cingulate cortex was decreased. Additionally, the study found a positive correlation between LC to right temporoparietal junction connectivity and MMSE scores in MCI patients, as well as hyperactivation of the left-insula salience network.

Summary: Sleep disruption is associated with cognitive impairment and the development of Alzheimer's disease. This study investigated the relationship between cognitive impairment non-dementia (CIND) and arousal indices (AIs) during different sleep stages. The results suggest that in CIND older adults, there is an attenuation of normal arousal mechanisms in REM sleep, possibly related to early pathological damage in the locus coeruleus noradrenergic system present in the Alzheimer's disease continuum.

Summary: This study used LC-MRI to investigate the involvement of the pontine nucleus Locus Coeruleus (LC) in Alzheimer's disease progression and its association with prognosis and cognitive performance in amnestic Mild Cognitive Impairment. The findings suggest that reductions in LC-MRI parameters may be predictive of clinical progression in Mild Cognitive Impairment and support the role of LC degeneration in the continuum of Alzheimer's disease.

Summary: This study investigates the relationship between noradrenaline (NA) depletion and the pathological expression of Tau and TDP-43 proteins, two major hallmarks of Alzheimer's disease (AD). The study suggests that the loss of NA is directly related to the phosphorylation of pathological Tau and TDP-43 proteins, which leads to working memory impairments. The findings indicate that enhancing noradrenergic function may have therapeutic potential in AD.

Summary: Adequate sleep is vital for maintaining health, but in modern society, with an accelerated pace of life, more people are sacrificing sleep to cope with social changes. Sleep deprivation has negative impacts on cognitive performance and overall health. However, moderate physical exercise has been found to prevent the detrimental effects of sleep deprivation through multiple pathways, as shown in this meta-analysis of 23 articles. This highlights the potential of exercise as a preventive or therapeutic strategy for sleep deprivation-induced negative effects.
