Parasites of the genus Plasmodium have a complex life cycle. They alternate between their final mosquito host and their intermediate hosts. The parasite can be either extra-or intracellular, depending on the stage of development. By modifying their shape, motility, and metabolic requirements, the parasite adapts to the different environments in their different hosts. The parasite has evolved to escape the multiple immune mechanisms in the host that try to block parasite development at the different stages of their development. In this article, we describe the mechanisms reported thus far that allow the Plasmodium parasite to evade innate and adaptive immune responses.

Summary: SARS-CoV-2 infection is generally mild, but in around 10% of cases, it can lead to severe pneumonia and potentially death. Genetic factors, such as inborn errors of type I interferons, and pre-existing auto-antibodies can increase the risk of critical illness. The production of type I interferons is essential for host defense against the virus.

Summary: In an international cohort of 112 hospitalized children with COVID-19 pneumonia, 12 children with recessive inborn errors of type I interferon immunity were identified. These deficiencies may contribute to the development of COVID-19 pneumonia in children. Additionally, these deficiencies were not found in individuals without pneumonia from SARS-CoV-2 infection.

Summary: Since the emergence of SARS-CoV-2 in December 2019, the COVID-19 pandemic has resulted in over 6.5 million deaths worldwide, highlighting the crucial role of the immune system in defending against emerging pathogens. Inborn errors of immunity (IEI), characterized by defects in immune cell development and function, make individuals highly susceptible to severe infections and immune dysregulation. The study of IEI has provided valuable insights into the mechanisms of disease pathogenesis and immunopathology, which can inform the development of therapies for both emerging and established infectious diseases.

Interfering with Interferons: A Critical Mechanism for Critical COVID-19 Pneumonia

Summary: Infection with SARS-CoV-2 can result in a wide range of clinical outcomes, from no symptoms to severe pneumonia and death. Genetic studies have shown that severe cases may be caused by defects in the type I interferon immune response or presence of autoantibodies that neutralize interferons. Virological and cellular studies have further demonstrated that SARS-CoV-2 proteins can hinder the induction or response to type I interferons, while these interferons can limit viral replication. These findings highlight the importance of sufficient type I interferon during the early stages of infection and have implications for treatment and future research.

Inherited and acquired errors of type I interferon immunity govern susceptibility to COVID-19 and multisystem inflammatory syndrome in children

Summary: Since the start of the COVID-19 pandemic, global sequencing efforts have made significant advancements in the field of inborn errors of immunity. Research has revealed that both known and novel inborn errors affecting type I interferon immunity play a critical role in severe COVID-19 cases, occurring in up to 5% of patients. Furthermore, autoantibodies against type I interferons have been found in around 20% of critically ill COVID-19 patients over 80 years old and 20% of fatal cases, with a higher prevalence in older males. Additionally, inborn errors impairing the regulation of type I interferon responses and RNA degradation have been identified as causes of multisystem inflammatory syndrome in children, which is a life-threatening condition that occurs following mild initial SARS-CoV-2 infection in children and young adults. Understanding these immunologic mechanisms can assist in the development of treatments for severe COVID-19, multisystem inflammatory syndrome in children, long COVID, and neuro-COVID.

In a study of COVID-19 patients and SARS-CoV-2naive individuals, it was found that autoantibodies targeting interferon-alpha were more common and associated with older age, while autoreactivity to interferon-gamma did not correlate with severe disease in COVID-19 patients.

Summary: Through a genome-wide rare variant burden association analysis, it was found that there is an association between at-risk variants in the TLR7 gene and rare loss-of-function variants in TLR3-dependent type I interferon immunity genes. These findings suggest that rare variants in TLR3- and TLR7-dependent type I interferon immunity genes may underlie life-threatening COVID-19 in patients under 60 years old.

Summary: The novel pyrrolizidinylmethyl derivative MG3 showed excellent antimalarial activity against drug-resistant parasites, both in vitro and in vivo. The compound exhibited comparable or better efficacy than chloroquine and other quinolines in rodent models of malaria, and its pharmacological profile met the requirements for further development.
