We explored the effects of sclerostin removal in vivo on experimental periodontitis in mice. A ligature of Porphyromonas gingivalis-saturated collagen silk was applied to the cervical region of the first molar tooth in 10 wild-type (WT) mice and 10 sclerostin-knockout (SOST-KO) mice, and the animals were fed 10% sucrose for 2 months. Another 10 WT mice and 10 SOST-KO mice were similarly treated, but then fed a normal diet for 2 months. The maxillae were then harvested for morphological and molecular examinations. The mice with periodontitis showed significantly more severe alveolar bone loss than control mice, the most significant absorption being observed in WT mice. Immunohistochemical staining demonstrated upregulation of RANKL and ERK1/2-MAPK expression and downregulation of OPG expression in mice with periodontitis, especially WT mice. Therefore, removal of sclerostin appears to modestly protect the alveolar bone from resorption in this experimental setting.

RANKL inhibition halts lesion progression and promotes bone remineralization in mice with fibrous dysplasia

Zhongyu Liu, Yijia Yin, Zheng Wang, Liang Xie, Peng Deng, Donghui Wang, Ning Ji, Hang Zhao, Xianglong Han, Qianming Chen, Chun-Hsi Chung, Ding Bai, Xuefeng Zhao

Summary: Fibrous dysplasia (FD) is a rare bone disease caused by GNAS mutation, and there is currently no cure available. Inhibition of RANKL expression has been identified as a potential therapy for FD. This study evaluated the effects of a small-molecule RANKL inhibitor (A52676293) on a transgenic FD mouse model, and found that it effectively suppressed osteoclastogenesis and halted FD progression. The results suggest that RANKL inhibition could be an effective treatment for FD.

Methods to assess tooth gingival thickness and diagnose gingival phenotypes: A systematic review

Jiangyue Wang, Sa Cha, Qing Zhao, Ding Bai

Summary: This systematic review aimed to explore the diagnostic accuracy of CBCT and ultrasound for gingival thickness measurement. The results showed no significant difference between CBCT measurement and transgingival probing in both anterior and posterior dentition. A moderate correlation was observed between CBCT and transgingival probing, while a weak correlation was found between ultrasound measurement and transgingival probing.

Heterostructured Metal-Organic Frameworks/Polydopamine Coating Endows Polyetheretherketone Implants with Multimodal Osteogenicity and Photoswitchable Disinfection

Yi Deng, Jiacheng Shi, Yau Kei Chan, Ding Bai, Rui Shu, Xiuyuan Shi, Yunfei Li, Limei Li, Xiao Yang, Weizhong Yang

Summary: In this study, a heterostructured coating was created on a porous bioinert polyetheretherketone implant. The heterostructured coating significantly promotes cytocompatibility and osteogenic differentiation through multiple mechanisms and effectively kills bacteria through photothermal/photodynamic effects and photo-induced accelerated ion release. The results confirmed the excellent osteogenicity and osseointegration of the heterostructured orthopedic implant, as well as its photoswitchable disinfection controlled by NIR light.

Effect of occlusal coverage depths on the precision of 3D-printed orthognathic surgical splints

Yipeng Wang, Peiqi Wang, Xiang Xiang, Hui Xu, Yuting Tang, Yumeng Zhou, Ding Bai, Chaoran Xue

Summary: The study proposed a standardized protocol for 3D-printed splints and assessed the precision of splints with different occlusal coverage depths. Results showed that deviations were more evident in the vertical dimension and pitch rotation, increasing as the occlusal coverage depth increased. Splints with 2-mm and 3-mm occlusal coverage depths are recommended for precision relative to clinical acceptability.

Clock-modified mesenchymal stromal cells therapy rescues molecular circadian oscillation and age-related bone loss via miR142-3p/Bmal1/YAP signaling axis

Sa Cha, Jiangyue Wang, Sueng Min Lee, Zhen Tan, Qing Zhao, Ding Bai

Summary: Age-related bone loss and disease have a significant impact on the quality of life for the elderly population. This study explores the role of cellular circadian rhythms and miRNAs in regulating bone aging. It shows that miR-142-3p downregulates Bmal1, reducing the osteogenic potential of BMSCs, while Bmal1 promotes BMSCs osteogenesis by inhibiting YAP expression. Cell-based circadian therapy improves bone formation and turnover levels.

Bin Li, Peiqi Wang, Hui Xu, Rui Gu, Xianglong Han, Ding Bai, Chaoran Xue

Summary: The study found that offset design influenced the accuracy of vertical bracket placement with GBDs, with TF being preferable to EF for GBDs due to smaller deviations in angulation and rotation. The differences between TF and EF also need to be considered in the design of other dental CAD/CAM devices.
