Summary: This study aimed to compare the SBAC-SI differences in AS, OCI, and DISH. The results showed that SBAC-SI was significantly higher in the OCI group compared to the AS group, which could help distinguish between these two diseases. In AS, the disease duration was negatively correlated with SBAC-SI, and HLA B27 was associated with lower SBAC-SI.

Summary: The aim of this study was to identify the role of Piezo1-mediated mechanotransduction in entheseal pathological new bone formation and explore the underlying molecular mechanism. It was found that Piezo1 was aberrantly upregulated in ligaments and entheseal tissues from patients with ankylosing spondylitis (AS) and animal models. Pharmacological and genetic inhibition of Piezo1 attenuated while its activation promoted pathological new bone formation. Mechanistically, activation of CaMKII signalling was found essential for Piezo1-mediated mechanotransduction. Additionally, Piezo1 was upregulated by AS-associated inflammatory cytokines.

Summary: Ankylosing spondylitis (AS) is a chronic inflammatory rheumatic disease that mainly affects the axial skeleton and is characterized by inflammatory back pain, bone structural damage, and pathological new bone formation. This study found increased purine metabolites in the plasma of AS patients and SHP2-deficient chondrocytes, suggesting the potential role of purinergic signaling in promoting ectopic new bone formation. Targeting purinergic signaling, specifically using the purinergic receptor antagonist Suramin, could potentially suppress pathological new bone formation in AS-like bone disease.

Results show significant reductions in arthritis, enthesitis, spondylitis, and ileitis in the mouse model treated with stat3-p lnh. In vitro experiments also indicate decreased frequencies of inflammatory cytokine-producing cells after stat3-p lnh treatment.

Summary: PDGFB plays a crucial role in the formation of enthesophytes in ankylosing spondylitis (AS) and is associated with spinal progression in AS. Increased levels of PDGFB were found in AS patients and correlated with radiographic progression. Inhibition of PDGFRB reduced PDGFB-induced bone mineralization and attenuated arthritis severity and enthesophyte formation in a mouse model. Regulating PDGFB/PDGFRB signaling could be a novel therapeutic strategy for AS.

Summary: In this study, manganese ferrite nanoparticles modified by the aptamer CH6 (CH6-MF NPs) were developed for the efficient treatment of ankylosing spondylitis (AS). The CH6-MF-Si NPs effectively suppressed abnormal osteogenic differentiation under inflammatory conditions in vitro and attenuated local inflammation and rescued heterotopic ossification in the entheses in a mouse model. These findings suggest that CH6-MF NPs may serve as an effective inflammation reliever and osteoblast-specific delivery system, and CH6-MF-Si NPs have potential for the dual treatment of chronic inflammation and heterotopic ossification in AS.
