The preadipocyte factor 1 (Pref-1) is involved in the proliferation and differentiation of various precursor cells. However, the intracellular signaling pathways that control these processes and the role of Pref-1 in the pancreas remain poorly understood. Here, we showed that Pref-1 induces insulin synthesis and secretion via two independent pathways. The overexpression of Pref-1 activated MAPK signaling, which induced nucleocytoplasmic translocation of FOXO1 and PDX1 and led to the differentiation of human pancreatic ductal cells into beta-like cells and an increase in insulin synthesis. Concurrently, Pref-1 activated Akt signaling and facilitated insulin secretion. A proteomics analysis identified the Rab43 GTPase-activating protein as a downstream target of Akt. A serial activation of both proteins induced various granular protein syntheses which led to enhanced glucose-stimulated insulin secretion. In a pancreatectomised diabetic animal model, exogenous Pref-1 improved glucose homeostasis by accelerating pancreatic ductal and beta-cell regeneration after injury. These data establish a novel role for Pref-1, opening the possibility of applying this molecule to the treatment of diabetes.

Cumulative Exposure to Metabolic Syndrome Components and the Risk of Dementia: A Nationwide Population-Based Study

Yunjung Cho, Kyungdo Han, Da Hye Kim, Yong-Moon Park, Kun-Ho Yoon, Mee Kyoung Kim, Seung-Hwan Lee

Summary: The study found that there is an association between cumulative exposure to metabolic disturbances, such as metabolic syndrome and its components, and the risk of dementia. The more exposure, the higher the risk, and even minimal exposure to metabolic disturbances can significantly impact the risk of dementia.

Summary: The study compared the non-clinical prerequisites for initiating clinical studies in five Asian countries/regions from an industry viewpoint. The findings revealed differences in regulations and development experiences, leading to variations in the non-clinical study package and its corresponding study design. Sharing experiences among Asian countries/regions could expedite cross-border IND approval and contribute to early delivery of innovative CTPs to patients.

Frequency of Exposure to Impaired Fasting Glucose and Risk of Mortality and Cardiovascular Outcomes

Seung-Hwan Lee, Kyungdo Han, Hyuk-Sang Kwon, Mee Kyoung Kim

Summary: The study found that the frequency and intensity of impaired fasting glucose (IFG) exposure are associated with cardiovascular disease and mortality risk. Adjusting for hypertension and dyslipidemia attenuated the association between high IFG exposure scores and risk of myocardial infarction or stroke, but the risk of all-cause mortality remained significant.

Retrospective cohort analysis comparing changes in blood glucose level and body composition according to changes in thyroid-stimulating hormone level

Hyunah Kim, Da Young Jung, Seung-Hwan Lee, Jae-Hyoung Cho, Hyeon Woo Yim, Hun-Sung Kim

Summary: The impact of thyroid-stimulating hormone (TSH) levels on diabetes mellitus incidence remains controversial in euthyroid state, but increases in TSH levels were associated with decreased skeletal muscle mass and increased body fat mass. Moreover, increment in TSH levels correlated with significant increases in fasting blood glucose and body mass index.
