Vitamin k3 inhibits protein aggregation: Implication in the treatment of amyloid diseases

Protein misfolding and aggregation have been associated with several human diseases such as Alzheimer's, Parkinson's and familial amyloid polyneuropathy etc. In this study, anti-fibrillation activity of vitamin k3 and its effect on the kinetics of amyloid formation of hen egg white lysozyme (HEWL) and A beta-42 peptide were investigated. Here, in combination with Thioflavin T (ThT) fluorescence assay, circular dichroism (CD), transmission electron microscopy and cell cytotoxicity assay, we demonstrated that vitamin k3 significantly inhibits fibril formation as well as the inhibitory effect is dose dependent manner. Our experimental studies inferred that vitamin k3 exert its neuro protective effect against amyloid induced cytotoxicity through concerted pathway, modifying the aggregation formation towards formation of nontoxic aggregates. Molecular docking demonstrated that vitamin k3 mediated inhibition of HEWL and A beta-42 fibrillogenesis may be initiated by interacting with proteolytic resistant and aggregation prone regions respectively. This work would provide an insight into the mechanism of protein aggregation inhibition by vitamin k3; pave the way for discovery of other small molecules that may exert similar effect against amyloid formation and its associated neurodegenerative diseases.

Summary: Microbial lipases are excellent biocatalysts that can catalyze various reactions in both aqueous and non-aqueous media. This review focuses on microbial lipases from different sources (psychrophiles, mesophiles, and thermophiles), highlighting the structural diversity in temperature adaptation and industrial applications. Understanding the molecular mechanisms of temperature adaptation in lipases is of great interest, as extremophilic lipases are widely used in biotransformation reactions with higher yields and unique products.

Binding of Novobiocin Paves the Way for Inhibition of DEAH-box Helicase 8

Summary: In this study, the interaction between human DHX8 and four test compounds (Etoposide, Netropsin, Nogalamycin, and Novobiocin) was investigated using molecular docking and fluorescence emission spectra techniques. The results showed a strong binding preference of DHX8 for Novobiocin, and a concentration of 2.5 μM of Novobiocin was found to effectively inhibit the activity of DHX8. These findings provide valuable insights into the inhibition of DHX8 and contribute to the development of Novobiocin as a therapeutic molecule against targeted diseases.

Summary: This study focuses on evaluating the anti-inflammatory and antioxidant effects of Bacterial Protease (BP) and 4,4'-(2,5-dimethoxy-1,4-phenylene) bis (1-(3-ethoxy phenyl)-1H-1,2,3-triazole) (DPET) in treating early-stage liver fibrosis induced by thioacetamide (TAA). The results showed that treatment with BP can improve the severe effects of TAA on the liver, restore tissue architecture, and have the potential to alleviate liver fibrosis.

Protein aggregation: Consequences, mechanism, characterization and inhibitory strategies

Summary: Proteins are vital for cellular functions, but variations in environmental conditions can disrupt their stability and lead to aggregation. Under normal conditions, aggregated proteins are removed by a quality control system, but diseased conditions or impaired systems can result in toxicity. Misfolding and aggregation of certain proteins are linked to diseases like Alzheimer's and Parkinson's. Current treatments are symptomatic and lack the ability to target the cause of these diseases. Therefore, more research is needed to develop drugs that address the root cause. This review provides valuable knowledge and strategies for researchers in neuroscience.

Summary: The aggregation of soluble proteins into insoluble fibrils is a key factor in the development of amyloid-associated disorders. In this study, the anti-diabetic drug chlorpropamide was found to effectively inhibit the aggregation of human lysozyme in vitro, reducing its cytotoxicity. Additionally, chlorpropamide was found to not only inhibit fibrillation but also stabilize the protein in its native state.
