17beta-estradiol counteracts neuropathic pain: a behavioural, immunohistochemical, and proteomic investigation on sex-related differences in mice

Sex differences play a role in pain sensitivity, efficacy of analgesic drugs and prevalence of neuropathic pain, even if the underlying mechanisms are far from being understood. We demonstrate that male and female mice react differently to structural and functional changes induced by sciatic nerve ligature, used as model of neuropathic pain. Male mice show a gradual decrease of allodynia and a complete recovery while, in females, allodynia and gliosis are still present four months after neuropathy induction. Administration of 17 beta-estradiol is able to significantly attenuate this difference, reducing allodynia and inducing a complete recovery also in female mice. Parallel to pain attenuation, 17 beta-estradiol treated-mice show a functional improvement of the injured limb, a faster regenerative process of the peripheral nerve and a decreased neuropathy-induced gliosis. These results indicate beneficial effects of 17 beta-estradiol on neuropathic pain and neuronal regeneration and focuses on the importance of considering gonadal hormones also in clinical studies.

Summary: This study identified gender differences in immune responses to neuropathic pain, with females showing higher infiltration of T cells in nerves and males showing higher infiltration of macrophages. Additionally, cytokine and chemokine levels were differently regulated in blood and nerve lysates from female mice compared to male mice. These findings provide insights into gender-associated inflammatory profiles in neuropathy and could contribute to the development of differential biomarkers and personalized medicine.

Summary: Amplification of the proto-oncogene MYCN is a key molecular aberration in high-risk neuroblastoma, affecting the protein cargo of extracellular vesicles (EVs) secreted by tumour cells and influencing the Warburg effect, proliferation, and c-MYC expression in recipient cells. The cancer-promoting activity of EVs was linked to the glycolytic kinase pyruvate kinase M2 (PKM2) enriched in EVs secreted by MYC-expressing neuroblastoma cells. Glycolytic enzymes PKM2 and hexokinase II were detected in EVs circulating in neuroblastoma patients but not in non-cancer children, indicating potential spread of oncogenic signals via EVs from MYC-activated cancers.

Summary: Glutathionylation and cysteinylation can protect critical cysteines from oxidative damages. Levels of S100A9 long and cystatin B in saliva were determined in preterm and at-term newborns, showing that the percentage of S-thiolated derivatives increased with postconceptional age, with the most significant variation occurring around 280 days. Differences in S-thiolated derivatives levels were found to be dependent on postconceptional age, not on preterm or at-term birth.

PPAR ligands, a class of drugs that activate Peroxisome Proliferator-Activated Receptors, were found to inhibit NADH coenzyme Q reductase and induce complex mitochondrial oxidative metabolism damage. This damage affects not only ATP synthesis but also respiratory control ratio, spare respiratory capacity, coupling efficiency, oxidative stress, and mitochondrial structural damage.

Botulinum Neurotoxins beyond Neurons: Interplay with Glial Cells

Summary: In recent years, studies have shown the significant therapeutic use of botulinum neurotoxins (BoNTs) in various motor and autonomic disorders. BoNTs exert their therapeutic effects by selectively cleaving specific sites of the SNARE protein complex, interfering with neural transmission. They can block the release of neurotransmitters and neuromodulators and affect glial cell function. The exact mechanisms are not fully understood, but BoNTs can inhibit spinal glial cell activation and promote peripheral regeneration. Overall, BoNTs have important therapeutic implications for conditions involving glial cell activation.

Role of Running-Activated Neural Stem Cells in the Anatomical and Functional Recovery after Traumatic Brain Injury in p21 Knock-Out Mice

Summary: Traumatic brain injury (TBI) is a common cause of death and disability worldwide, with no effective treatments available. Recent studies have suggested that adult neurogenesis may be a new approach to counteract post-traumatic neurodegeneration. It has been shown that long-term running can enhance adult neurogenesis and promote neuroregeneration and functional recovery after injury. These findings suggest that running could be a promising method to increase neural stem cells and improve endogenous neuroregenerative responses following brain trauma.
