Summary: This article presents an integrative review on the implications of semaphorins in cancer, specifically discussing their role in cell proliferation/survival, angiogenesis, invasion, metastasis, stemness, and chemo-resistance/response. It highlights the heterogeneity of semaphorins as a family and their interaction with the tumor microenvironment components in driving cancer progression. The differences observed in the biological structure of semaphorins and the specificities of each cancer type necessitate viewing each semaphorin as a unique entity and conducting research accordingly. A deeper understanding of the molecular mechanisms promoting and sustaining the malignant behavior of cancer cells is crucial.

Summary: RNA methylation, including m6A, m5C, and m1A, plays a critical role in cancer. Methylation-associated lncRNAs are involved in various biological processes in cancer, and our analysis identified 39 lncRNAs strongly associated with prognosis in pancreatic cancer. Using a combination of clinical characteristics, we developed a risk model comprising seven lncRNAs that accurately predicted the survival probability of patients. This study highlights the potential of m6A/m5C/m1A-associated lncRNAs as biomarkers for early diagnosis, prognosis estimation, and response prediction to immunotherapy in pancreatic cancer patients.

Summary: This study aimed to investigate the effects of androgen receptor signaling pathway-related long non-coding RNAs (ARSP-related lncRNAs) on breast cancer subtype classification and tumor microenvironment (TME). By screening ARSP-related lncRNAs, a risk model was constructed and found to be an independent prognostic factor for breast cancer patients. The risk score also had an impact on immune cell infiltration in the TME.

Summary: This study found that in colorectal cancer (CRC), the expression of FOXS1 is upregulated and is positively correlated with poor survival. FOXS1 promotes malignant behavior of CRC cells, including proliferation, invasion, and angiogenesis, both in vitro and in vivo. Mechanistically, FOXS1 upregulates the transcriptional level of CXCL8 to regulate tumor growth and metastasis. These findings suggest that the FOXS1/CXCL8 axis may serve as a potential therapeutic target for the treatment of metastatic CRC.

Summary: Insulin resistance in the heart is a characteristic of diabetic cardiac injury. The molecular mechanisms underlying this resistance are not yet understood. Recent studies have shown that the diabetic heart is also resistant to other cardioprotective interventions. The role of Cav3 in diabetic impairment of cardiac protective signaling and diabetic ischemic heart failure needs further investigation.
