Summary: Liquid biopsy, specifically plasma ctDNA analysis, has revolutionized cancer patient management by detecting genomic alterations and resistance mechanisms, particularly in lung cancer patients. While offering a comprehensive view of tumor diversity, its non-invasive nature overcomes the challenges of obtaining tissue samples. Further clarification is needed regarding the use of LB in supporting diagnostic and therapeutic decisions.

Summary: Compared to earlier stages, stage IV gastric cancer patients showed significantly higher levels of cell-free DNA (cfDNA). This study analyzed cfDNA and tumor DNA using next-generation sequencing (NGS) and found different mutation patterns between the two. The most commonly mutated genes in tumor samples were TP53, ARID1A, KMT2C, and KMT2D, while in cfDNA samples, FAT4 and MACF1 were the most commonly mutated genes. The concordance of mutation patterns between cfDNA and tumor DNA was 42.0%. The study suggests that monitoring cfDNA mutation patterns could be useful in the treatment of stage IV gastric cancer.

Summary: This pilot study demonstrated that combining CTC cluster counts and cfDNA levels can predict progression-free survival in NSCLC patients. The results encourage further exploration of the combined effect of CTC/cfDNA as a prognostic biomarker in a large cohort of advanced stage NSCLC patients.

Summary: Recent research suggests that changes in ctDNA levels are correlated with TB changes, and the detection of ctDNA can be used to predict new metastases, rapid tumor growth, and poor survival in NSCLC patients.

Summary: This study developed a comprehensive cfRNA-Seq pipeline and a machine learning model to assist in the non-invasive early diagnosis of NSCLC. The results identified differential mRNA, lncRNAs, and miRNAs from cfRNA-Seq that were significantly associated with the development and progression of lung cancer. The classifier based on gene expression signatures showed high specificity and sensitivity. Additionally, the analysis of T cell and B cell immune repertoire and the microbiome analysis provided insights into the immune status and microbial profiles of NSCLC patients.

Summary: The study demonstrated that the STAT3/ZEB1 axis is critical in gefitinib resistance in lung cancer, and a new potential therapeutic strategy targeting STAT3 has been identified with the inhibitor LL1. LL1 was shown to sensitize resistant cells to gefitinib by depleting STAT3 activity and blocking its signaling pathways, with little observed toxicity in animal models, indicating it could be a chemotherapeutic adjuvant for gefitinib resistance in NSCLC.

Summary: The study demonstrated that a high RS was associated with a higher objective response rate, longer progression-free survival, and overall survival. Changes in variant allele frequency (VAF) after two cycles of chemotherapy could predict treatment response. Monitoring VAF levels could provide valuable information for assessing treatment efficacy.

Summary: This study analyzed the levels of circulating dd-cfDNA in different types of graft rejection and evaluated its diagnostic potential in predicting graft rejection after lung transplantation. The results showed that circulating dd-cfDNA levels were significantly higher in graft rejection, ACR, and AMR compared to non-rejection. Circulating dd-cfDNA could be used as a non-invasive biomarker to distinguish patients with graft rejection from normal stable controls.

Summary: This study presents a novel method for in situ isolating and directly detecting circulating tumor cells (CTCs) from peripheral blood using black TiO2-based Surface-Enhanced Raman Scattering (SERS) bio-probe. The strategy showed high efficiency, reliability, and specificity in distinguishing CTCs from peripheral blood cells. It significantly reduced the detection time and improved the detection sensitivity, making it a potential tool for liquid biopsy.

Summary: Lung cancer is a leading cause of cancer-related death, and early diagnosis is crucial. Plasma circulating cell-free DNA (cfDNA) can be used non-invasively and cost-effectively to detect lung cancer through high-sensitivity sequencing techniques, as it contains comprehensive genetic and epigenetic information from tissues throughout the body.

Summary: The study successfully identified a specific combination of six metabolic biomarkers that enables discrimination between stage I lung cancer patients and healthy individuals, and proposed Naive Bayes as an exploitable tool for early lung tumor prediction. The research provides strong support for the feasibility of blood-based screening for lung cancer, and the interdisciplinary method could potentially be adapted to other cancers beyond lung cancer.

Summary: This study retrospectively analyzed the diagnosis and treatment course of 14 cases of exudative pleural effusion (EPE) caused by lung fluke infection and proposed a practical diagnosis approach. It was found that all patients had eosinophilia in peripheral blood and a history of special ingestion. Lung fluke infection should be highly suspected in patients with unexplained pleural effusion, especially when pleural fluid or pleural biopsy shows eosinophilic PE or eosinophils infiltration.
