Coumarins from the roots of Angelica dahurica with antioxidant and antiproliferative activities

Angelica dahurica has been frequently used as a food additive and a folk medicinal herb in Asian countries. The aim of this study was to isolate key active compounds from A. dahurica with antioxidant and anticancer activities. Three new furocoumarin dimers and twenty known coumarins were obtained from A. dahurica. Their structures were identified by extensive 1D and 2D NMR, CD, and HR-ESIMS spectroscopic analyses and screened by UPLC-MS/MS. Imperatorin oxypeucedanin hydrate, xanthotoxol, bergaptol, and 5-methoxy-8-hydroxypsoralen exhibited moderate DPPH center dot scavenging activity and strong ABTS(center dot+) scavenging activity. Isoimperatorin, phelloptorin, and pabularinone showed significant inhibition on HepG2 cells with IC50 values of 8.19, 7.49, and 7.46 mu M, respectively. Furthermore, pabularinone also showed moderate inhibition on HeLa cells with an IC50 value of 13.48 mu M. These results suggested that A. dahurica could be explored as new and potential natural antioxidants and cancer prevention agents for use in functional foods. (C) 2015 Elsevier Ltd. The latest advances in small-molecule fluorescent probes for nitroreductase detection and the development trends in this field were summarized in this review.

Discovery of a Novel Vascular Disrupting Agent Inhibiting Tubulin Polymerization and HDACs with Potent Antitumor Effects

Summary: A series of novel multifunctional vascular disrupting agents (VDAs) was designed and synthesized, among which compound TH-6 showed the most potent antiproliferative activity against a panel of cancer cell lines. TH-6 inhibited tubulin assembly and increased the acetylation level of HDAC substrate proteins, leading to effective tumor growth inhibition without apparent toxicity. Furthermore, TH-6 disrupted both the internal and peripheral tumor vasculatures, contributing to persistent tumor inhibitory effects. Therefore, TH-6 deserves further investigation for its potential as a new approach in clinical cancer therapy.

Abietane diterpenes from the twigs and leaves of Cephalotaxus oliveri Mast. with antitumor activity

Summary: Twenty one abietane diterpenes were isolated from Cephalotaxus oliveri Mast, including 7 undescribed diterpenoids named cephaloliverins A-G. Metaglyptin A exhibited the strongest cytotoxicity against three human tumor cell lines (MCF-7, HepG2, and A549) and further investigations were conducted on its effects on colony formation and apoptosis in HepG2 cells.

Flavonolignans and biflavonoids from Cephalotaxus oliveri exert neuroprotective effect via Nrf2/ARE pathway

Summary: Plants of the Cephalotaxus genus, including Cephalotaxus oliveri, contain diverse and bioactive components. In this study, several undescribed flavonolignans and biflavonoids were isolated from C. oliveri, along with known compounds. The isolated compounds showed neuroprotective activity against H2O2-induced cell injury and umcephabiflovin B demonstrated the strongest effect.

Design and Synthesis of Brefeldin A-Isothiocyanate Derivatives with Selectivity and Their Potential for Cervical Cancer Therapy

Summary: A total of 25 brefeldin A-isothiocyanate derivatives were synthesized and tested. Most derivatives showed selectivity between HeLa and L-02 cells. In particular, derivative 6 exhibited potent antiproliferative activity against HeLa cells with no cytotoxic effects on L-02 cells. Mechanism studies revealed that derivative 6 induced cell cycle arrest at G1 phase and apoptosis through the mitochondrial-dependent pathway.

A photoactivated H2S donor based on a coumarin structure with real-time monitoring capability

Summary: We developed a photoactivated H2S donor, Bhc-TCN-Ph, derived from 6-bromo-7-hydroxycoumarinmethyl thiocarbonate. COS is released upon 365nm light exposure, generating H2S and coumarin fluorescence for visualization. This process does not produce electrophilic by-products. In vitro evaluations demonstrate good cytochemical and cytocompatibility characteristics.

Cephaloliverols A and B, two sterol-hybrid meroterpenoids from Cephalotaxus oliveri

Summary: Cephaloliverols A and B, two new meroterpenoids based on a sterol and an abietane diterpene, were isolated from the twigs and leaves of Cephatotaxus oliveri. Their structures were determined by spectroscopic analysis and quantum chemical calculation. The compounds and their precursors showed a dose-dependent decrease in NO production in LPS-stimulated RAW 264.7 macrophages.
