Summary: Intraovarian growth factors, such as transforming growth factor-alpha (TGF-α), play a crucial role in determining the fate of ovarian follicles. TGF-α stimulates granulosa cell proliferation in small antral follicles through MAPK/ERK signaling pathway. However, it does not prevent apoptosis, suggesting that TGF-α may not be able to promote the persistence of follicles during follicular selection/dominance. Understanding the effects of TGF-α on granulosa cell function is essential for unraveling the mechanisms of ovarian follicle development and fertility in animals.

Summary: The aim of this study was to improve the in vitro culture of bovine oocytes collected from early antral follicles (EAFs) by maintaining physiological conditions and adding key hormones. The culture system involved a 5-day culture period and 3D-like structures. The cultured oocytes showed morpho-functional changes, improved developmental competence, and potential for in vitro embryo production.

Summary: This study identified the potential role of the Pde4d gene in polycystic ovary syndrome. Experimental validation showed that overexpression of Pde4d led to the formation of ovarian cysts and inhibited follicle maturation and apoptosis. This finding is important for understanding the pathogenesis of polycystic ovary syndrome.

Summary: AMH, a member of the TGF-beta superfamily produced by follicular granulosa cells in women, is thought to inhibit aromatase and play a role in ovarian steroidogenesis. This study found a negative correlation between follicle diameter and AMH forms, and variability in AMH isoforms between follicles of identical size within the same woman. The complex intrafollicular processing of AMH may pose challenges for developing an antibody-based AMH assay.

Summary: In this study, single-cell RNA sequencing was used to explore the characteristics of porcine antral follicles. The analysis revealed distinct subpopulations within mural and cumulus granulosa cells, indicating diverse functions within the follicles. Functional enrichment analysis identified specific subpopulations involved in steroid biosynthesis, cumulus expansion, and cellular communication. Comparing mature and less mature follicles highlighted developmental-specific variations in cell distribution and functions. These findings shed light on the intricate cellular heterogeneity and communication network within porcine antral follicles, providing valuable insights into the regulation of follicle development and oocyte maturation. These results hold promise for improving pig reproductive efficiency and advancing human reproductive medicine.

Summary: A study found that treatment with high dose of growth hormone (GH) can promote the response to superovulation and improve the developmental ability of oocytes in prepubertal lambs, resulting in an increase in the production of high-quality embryos. Additionally, GH also regulates ovarian function by altering the abundance of metabolites in follicular fluid, such as antioxidants and fatty acids.

Summary: This study used high-throughput RNA sequencing to identify differentially expressed genes in granulosa cells from healthy and early atretic ovarian follicles in pigs. GO analysis highlighted hormone metabolism, plasma membrane localization, and transporter activity, while pathway analysis revealed the involvement of steroidogenesis, cell adhesion molecules, and TGF-beta signaling pathways in atresia.

Summary: The study evaluated the proteome of early antral follicles from Ovis aries using LC-MS/MS analysis. A total of 2503 follicle proteins were identified, including abundant proteins such as vimentin, actin, lamin, heat shock proteins, and histones. miRNAs were found to modulate genes involved in cell cycle, cell differentiation, aging, apoptosis, adipocyte differentiation, and cell division in sheep follicles. This comprehensive atlas of proteins expressed in sheep early antral follicles will contribute to the identification of biomarkers for follicular development and oocyte maturation.

Summary: This study characterized the glucose and redox metabolism in mouse antral follicles with meiotically blocked oocytes after in vitro follicle culture (IFC). The results showed that granulosa cells in the blocked follicles had higher levels of glycolysis and lactic acid fermentation, while oocytes had increased activity in the pentose phosphate pathway.
