Summary: This study evaluated the new bone formation potential of MBCP and Bio-Oss grafting materials with DPSCs and BMSCs in a rabbit calvarial bone defect model. The results demonstrated that autogenous bone is the gold standard, and both DPSCs and BMSCs enhanced the osteoconductive capacities of MBCP and Bio-Oss. Moreover, the efficiency of BMSCs combined with MBCP and Bio-Oss was comparable to that of autogenous bone after 8 weeks of healing.

Summary: Teeth contain mesenchymal stem cells (MSCs) that promote tooth growth and repair. Dental-derived stem cells (d-DSCs), including dental pulp stem cells (DPSCs) and dental bud stem cells (DBSCs), have shown great potential in bone tissue engineering. The use of bone-associated factors and small molecular compounds has been found to enhance stem cell differentiation and osteogenesis. Natural and non-natural compounds found in fruits, vegetables, and drugs have also been explored for their ability to enhance MSC osteogenic differentiation and bone formation. This review focuses on the research conducted in the past decade on DPSCs and DBSCs, aiming to identify compounds that can stimulate d-DSC proliferation and osteogenic differentiation for bone regeneration.

This review focuses on the effects of epigenetic regulation on osteogenic differentiation, proliferation, and cellular senescence in MSCs and its role in bone regeneration.

Summary: Nerve growth factor (NGF) is known for its role in nervous system development and cell survival/differentiation. Recent studies have found that NGF also enhances bone regeneration. This study shows that NGF mediates osteogenic differentiation through the p75(NTR) pathway, activating the JNK cascade and KDM4B expression to remove repressive epigenetic marks and promote the expression of DLX5, a master osteogenic gene. KDM4B and c-Jun from the JNK signaling pathway work together to regulate NGF-mediated osteogenic differentiation.

Summary: In this study, a rabbit model was established to mimic human osteonecrosis of the femoral head (ONFH) and evaluate the autologous MSC-based treatment. Results showed that implantation of autologous BM-MSCs could restore the damaged tissue in rabbit femurs. Furthermore, the study found that osteonecrosis did not affect the behavior of MSCs.
