Determination of a CD4+CD25-FoxP3+ T cells subset in tumor-draining lymph nodes of colorectal cancer secreting IL-2 and IFN-γ

CD4(+)CD25(-)FoxP3(+) cells are a newly recognized subset of T cells which was first reported in autoimmune diseases. In our previous study, this subset was detected in tumor-draining lymph nodes (TDLNs) of patients with breast cancer. As little is known about their function in TDLNs of cancer patients, in this study, their frequency as well as their ability to produce interleukin (IL)-2, IL-10, or interferon (IFN)-gamma were investigated in TDLNs of colorectal cancer (CRC) patients. Mononuclear cells were isolated from lymph nodes of 13 patients with CRC using Ficoll-Hypaque gradient centrifugation. Cells were stimulated in vitro and stained with CD25, CD4, FoxP3, IFN-gamma, IL-10, and IL-2 or isotype matched antibodies and subjected to flow cytometry. The frequency of CD4(+)CD25(-)FoxP3(+)CD127(dim/-) cells was significantly lower than CD4(+)CD25(+)FoxP3(+)CD127(dim/-) population in TDLNs of CRC patients. The percentage of CD127(dim/-) cells and also the MFI of FoxP3 expression was significantly lower in CD4(+)CD25(-)FoxP3(+) in comparison with CD4(+)CD25(+)FoxP3(+) population. Moreover, CD4(+)CD25(-)FoxP3(+) cells contained higher percentages of IL-2- and IFN-gamma-producing cells than CD4(+)CD25(+)FoxP3(+) subpopulation. But, no difference was seen between two subsets in terms of IL-10 production. CD4(+)CD25(-)FoxP3(+) cells in TDLNs of CRC patients had lower suppressive and higher effector properties in comparison with CD4(+)CD25(+)FoxP3(+) conventional regulatory T cells.

The mTOR pathway plays an important role in viral replication, and mTOR pathway inhibitors may be a potential approach to anti-SARS-CoV-2 therapy.

The expression pattern of membranous TNF-? is distinct from its intracellular form in breast cancer-draining lymph nodes

Summary: This study focused on the expression of membranous and intracellular forms of TNF-alpha in lymphocytes derived from breast cancer-draining lymph nodes. The results showed that CD4+T cells primarily express the membranous form, while CD8+T cells primarily express the intracellular form. The intensity of membranous TNF-alpha expression in CD19+ cells was correlated with poor prognosticators, and the frequency of membranous TNF-alpha+CD19+ cells showed an inverse correlation with the number of involved lymph nodes.

Summary: Urothelial bladder cancer (UBC) is a complex malignancy with a high recurrence rate. Current diagnostic procedures lack sensitivity and specificity, thus biomarkers have potential value for early diagnosis, prognosis, and immunotherapy. Proteomics methods have rapidly advanced in identifying UBC biomarkers, and a panel of biomarkers may achieve higher diagnostic accuracy. This review evaluates recent literature on UBC proteomics, classifies tumor biomarkers based on their sources, and discusses the limitations and advantages of each source.

Quercetin potentiates the anti-osteoporotic effects of alendronate through modulation of autophagy and apoptosis mechanisms in ovariectomy-induced bone loss rat model

Summary: The study investigates the combined effects of quercetin and alendronate on osteoporosis markers, autophagy, and apoptosis in ovariectomized rats. The results show that quercetin enhances the anti-osteoporotic effects of alendronate, possibly through the regulation of autophagy and apoptosis mechanisms. The combination of quercetin and alendronate could be a useful therapeutic strategy in the treatment of osteoporosis in postmenopausal women.

Effect of Disease-Modifying Therapies on COVID-19 Vaccination Efficacy in Multiple Sclerosis Patients: A Comprehensive Review

Summary: According to current knowledge, the etiology of multiple sclerosis (MS) is complex and involves both genetic and environmental factors. MS is an immune-mediated, inflammatory neurological disease that results in myelin and axonal loss. There are approximately 20 disease-modifying therapies (DMTs) available that reduce disease activity in MS patients. Since MS patients have a high rate of disability and increased susceptibility to infections, the outbreak of COVID-19 poses a significant concern. Additionally, the immunosuppressive properties of DMTs during the pandemic raise questions about their impact on COVID-19 risks and vaccination efficacy in MS patients.

Production of a biosimilar version of aflibercept to improve VEGF blocker cytotoxicity on endothelial cells

Summary: This study aimed to produce a biosimilar version of aflibercept (AFL) and evaluate its effect when used in combination with other vascular endothelial growth factor (VEGF) blocker drugs. The biosimilar-AFL showed significant inhibition potential on HUVEC cells in a dose-dependent manner. Furthermore, co-treatment with Everolimus (EVR), Lenvatinib (LEN), and Sorafenib (SOR) enhanced the cytotoxicity of biosimilar-AFL on HUVEC cells, with LEN and SOR showing a 10-fold increase in cytotoxicity. The combination of biosimilar-AFL with LEN showed the most efficient result, while the combination with EVR showed the least efficient result. Overall, biosimilar-AFL may improve the efficacy of LEN, EVR, and SOR in reducing the VEGF effect on endothelial cells.

Comparative proteomics analysis in different stages of urothelial bladder cancer for identification of potential biomarkers: highlighted role for antioxidant activity

Summary: The aim of this study is to recognize tumor-associated proteins, which are differentially expressed in different stages of UBC patients compared to non-cancerous tissues. The study found significant changes in protein expression related to oxidative stress and antioxidant activity in UBC patients. These findings may contribute to the development of promising biomarkers for early detection, monitoring, and prognosis of UBC.

Epidemiological Profile of Salivary Gland Tumors in Southern Iranian Population: A Retrospective Study of 405 Cases
