Summary: This article describes the process of generating a CD34+ cell product as a starting material to produce HLA-homozygous iPSC following a cost-effective and clinical grade-compliant procedure, starting from unit selection and donor reconsenting. These CD34+ cells serve as the basis for the Spanish bank of haplolines, aimed to provide cell products for clinical research and therapy.

Summary: HPL serves as a valuable alternative to fetal bovine serum in cell culture, allowing for efficient and animal serum-free cell propagation. Providers must adhere to strict quality control and regulatory requirements to ensure safe and GMP-compliant production of HPL. The starting material for HPL preparation is fresh or expired platelet concentrates from healthy donors, and the process may involve pathogen reduction technologies.

Summary: Allogeneic natural killer (NK) cell-based immunotherapy is a promising approach for acute myeloid leukemia (AML). A good manufacturing practice (GMP)-compliant, cytokine-based ex vivo manufacturing process for generating NK cells from CD34(+) hematopoietic stem and progenitor cells (HSPC) has been developed. This process generates a consistent and active NK cell product that meets the requirements for NK cell immunotherapy. The manufactured NK cell batches are used in a clinical trial approved by the Dutch Ethics Committee.

Summary: Human embryonic stem cells (hESCs) are being used more and more in clinical trials due to their potential to improve the outcomes of treatment for various human diseases. Ensuring the quality of hESCs is crucial, and current good manufacturing practice (cGMP) guidelines are followed during the transport of embryos and the derivation of hESC cultures. This paper presents the derivation of three clinical-grade hESC lines that meet these criteria and demonstrates the development and validation of a system for manufacturing high-quality starting materials suitable for cell therapy according to cGMP.

Summary: This study demonstrates that the use of HPL-S is an effective alternative for MSC production under GMP conditions. The protein and growth factor content in HPL-S is comparable to that of HPL-E. Furthermore, MSCs derived from HPL-S show lower HLA-DR expression in immunophenotypic analysis.

Summary: This article introduces the establishment of clinical-grade human embryonic stem cell lines KthES11 in Japan according to good manufacturing practice standards. By using xeno-free chemically defined medium and optimizing the protocol, they successfully established five hESC lines, which are of great significance for human pluripotent stem cell therapy applications.

Summary: This study focuses on stochastic flow lines with limited material supply, where shortages of material supply can limit system performance. By using linear programming approaches, the impact of material shortages on system output is quantified, and optimal material levels are determined to prevent shortages and optimize system performance. Insights are gained on the accuracy of linear programs and the dependence of optimal material levels on flow line characteristics.

Summary: The current evaluation of the building industry reveals low efficiency in material use and automation level, as well as a scarcity of skilled workers in mechanical engineering and construction. Therefore, there is a noticeable shift towards automated production processes, particularly in the field of precast concrete construction. The high innovation spirit in individual research facilities and companies has led to various new applications of robotics and automated systems in the precast industry.

Summary: This study aimed to generate transplantable midbrain dopaminergic (mDA) progenitors for treatment of Parkinson's Disease (PD). By optimizing the differentiation protocol using clinical-grade human embryonic stem cells (hESC), the researchers successfully obtained functional mDA progenitors. Transplantation of cryopreserved mDA progenitors into rats with 6-hydroxydopamine lesions resulted in functional recovery. This study provides a foundation for future translational applications.
