Summary: The emergence of cell and gene therapies has sparked interest, but due to high manufacturing costs and long development times, there is a need for more efficient platforms. By improving the design of the stirred-tank bioreactor, suspension of microcarriers/beads for cell culture has been enhanced, showing promise for future applications in cell therapy and gene therapy.

Role of extracellular vesicles from adipose tissue- and bone marrow-mesenchymal stromal cells in endothelial proliferation and chondrogenesis

Summary: The secretome of mesenchymal stromal cells (MSCs) derived from different tissue sources is considered an innovative therapeutic tool for regenerative medicine. The study compared middle- and small-sized extracellular vesicles (EVs) released by adipose tissue-derived MSCs (ADSCs) and bone marrow-derived MSCs (BMSCs), showing different protein content and functional effects, with ADSC-EVs overexpressing pro-angiogenic factors and BMSC-EVs containing a higher amount of pro-differentiation and chemotactic proteins. This study highlights the importance of selecting the appropriate cell source of EVs for targeted therapeutic applications.

Summary: Advanced therapeutic medicinal products (ATMPs) have created a demand for large volumes of high-quality GMP cells. This study presents a fully automated, robot-assisted platform for scalable production of adherent human stem cells. The platform allows for a closed process from bone marrow collection to cell isolation, expansion, and collection into cryovials for patient delivery.

Regenerative and Anti-Inflammatory Potential of Regularly Fed, Starved Cells and Extracellular Vesicles In Vivo

Summary: This study evaluated the anti-inflammatory and regenerative effects of bone marrow MSC and their extracellular vesicles in a diabetic fractured mouse model. The results showed that preconditioned cells promoted earlier soft callus formation and mineralization at the fracture site, suggesting the importance of preconditioning in MSC therapy research and optimization.

Summary: The advent of immune checkpoint inhibitors (ICIs) has dramatically changed the treatment of non-small cell lung cancer (NSCLC). However, their efficacy remains limited to a subset of patients. This study evaluated the prognostic role of soluble (s) immune checkpoints and immune checkpoints associated with extracellular vesicles (EVs) in NSCLC patients treated with ICIs. The results showed that high levels of sPD-L1, sB7-H3, and sB7-H4 were associated with different prognoses in advanced NSCLC patients treated with ICIs. The study also suggested potential involvement of EVs in the disease response to ICIs.

Scalable manufacturing of gene-modified human mesenchymal stromal cells with microcarriers in spinner flasks

Summary: This study demonstrates a scalable manufacturing process using a microcarrier-bioreactor system for the expansion of gene-modified human mesenchymal stromal cells (hMSCs). The transduced and expanded cells met the criteria established by the International Society for Cell and Gene Therapy (ISCT). This work provides a foundation for the scalable manufacturing of gene-modified hMSCs.

Optimizing and developing a scalable, chemically defined, animal component-free lentiviral vector production process in a fixed-bed bioreactor

Summary: This study optimized and developed a scalable, chemically defined, animal component-free LVV production process using adherent human embryonic kidney 293T cells in a fixed-bed bioreactor. By changing the process conditions, such as reducing cell seeding density, delaying transient transfection, reducing plasmid DNA dosage, and adding compound, the LVV titer was improved by 26.9-fold. The results showed comparable LVV titers with traditional culture methods at different scales.

Pilot study investigating BP-180 in extracellular vesicles derived from blister fluid of bullous pemphigoid patients

Summary: This study aimed to investigate the presence of BP targeted autoantigens in blister fluid-derived extracellular vesicles (EVs). EVs were isolated from blister fluid of BP patients and suction blister fluid of healthy donors, and EV characterization was performed. Western blot analysis revealed the presence of BP180 in EVs derived from blister fluid of 3 out of 6 BP patients, while no BP180 was detected in EVs from healthy donors. This discovery provides insights into the pathogenesis of BP.
