The mountainous Cretan dietary patterns and their relationship with cardiovascular risk factors: the Hellenic Isolated Cohorts MANOLIS study

Objective: We carried out de novo recruitment of a population-based cohort (MANOLIS study) and describe the specific population, which displays interesting characteristics in terms of diet and health in old age, through deep phenotyping. Design: Cross-sectional study where anthropometric, biochemical and clinical measurements were taken in addition to interview-based completion of an extensive questionnaire on health and lifestyle parameters. Dietary patterns were derived through principal component analysis based on a validated FFQ. Setting: Geographically isolated Mylopotamos villages on Mount Idi, Crete, Greece. Subjects: Adults (n 1553). Results: Mean age of the participants was 61.6 years and 55.8% were women. Of the population, 82.7% were overweight or obese with a significantly different prevalence between overweight men and women (43.4 v. 34.7%, P = 0.002). The majority (70.6%) of participants were married, while a larger proportion of women were widowed than men (27.8 v. 3.5%, P<0.001). Smoking was more prevalent in men (38.7 v. 8.2%, P<0.001), as 88.8% of women had never smoked. Four dietary patterns emerged as characteristic of the population; these were termed 'local', 'high fat and sugar, 'Greek cafe/tavern' and 'olive oil, fruits and vegetables'. Individuals more adherent to the local dietary pattern presented higher blood glucose (beta = 4.026, P<0.001). Similarly, individuals with higher compliance with the Greek cafe/tavern pattern had higher waist-to-hip ratio (beta = 0.012, P<0.001), blood pressure (beta = 1.015, P=0.005) and cholesterol (beta = 5.398, P<0.001). Conclusions: Profiling of the MANOLIS elderly population identifies unique unhealthy dietary patterns that are associated with cardiometabolic indices.

Summary: Twin studies suggest shared genetic factors between eating disorders and substance use disorders, with strongest associations between bulimia nervosa and problem alcohol use. Genome-wide association studies reveal significant positive genetic correlations between alcohol use disorder and anorexia nervosa, cannabis initiation and anorexia nervosa, and cannabis initiation and anorexia nervosa with binge eating. Conversely, significant negative genetic correlations were observed between smoking behaviors and anorexia nervosa without binge eating.

Summary: Sex differences play a role in genetic effects on fasting glucose and insulin levels, with a sex dimorphism in allelic effects on fasting insulin and sex-homogeneous effects on fasting glucose. In addition, there are stronger genetic correlations between fasting insulin and waist-to-hip ratio/anorexia nervosa in women, and waist-to-hip ratio is causally related to insulin resistance in women but not in men. These findings highlight the importance of understanding metabolic and glycemic health sex dimorphism in exploring genetic effects in related phenotypes between women and men.

Construction and analysis of protein-protein interaction network of non-alcoholic fatty liver disease

Summary: Non-alcoholic fatty liver disease (NAFLD) is a complex disease that requires identification and study of new candidate proteins. This study analyzed the interaction network of human proteins associated with NAFLD to identify novel associations with other proteins involved in the disease. The computational analysis detected 77 candidate proteins with high network scores, and clustering analysis identified densely connected regions with biological significance. Additionally, gene expression analysis was conducted to validate some of the research findings, aiming to provide insights for addressing the pathogenesis and progression of NAFLD.

Risk Assessment for Hip and Knee Osteoarthritis Using Polygenic Risk Scores

Summary: This study examined the association between polygenic risk scores (PRS) and hip and knee osteoarthritis (OA) using data from population-based and clinical cohorts. The results showed a stronger association between PRS and clinically defined OA compared to radiographic OA phenotypes, with the highest risk stratification observed for total hip replacement (THR) or total knee replacement (TKR). These findings suggest that PRS-based risk assessment could play a role in preventing OA, but further studies are needed to test its integration in diverse healthcare settings.

Furthermore, it suggests that the effectiveness of certain dietary patterns on serum triglyceride levels may be influenced by specific genetic variations, particularly TM6SF2-rs58542926.

Associations of VEGF-A-Related Variants with Adolescent Cardiometabolic and Dietary Parameters

Summary: This study investigated the potential relationship between VEGF-A-related SNPs, cardiometabolic parameters, and dietary habits in adolescents. These findings provide the first-ever attempt to examine the influence of VEGF-A-related variants on teenage cardiometabolic determinants and reveal the modifying effect of diet.

Summary: This study aimed to investigate the relationship between MRI parameters reflecting inflammation and fibrosis and plasma amino acid (AA) concentrations in NAFLD patients. The study found that subjects with higher MRI-proton density fat fraction (MRI-PDFF) had higher plasma AA levels compared to those with lower PDFF. Significant associations were also observed between AAs and MRI-PDFF and MRI-cT1, suggesting the potential utility of circulating AAs as diagnostic markers for NAFLD.
