Liposomal systems as viable drug delivery technology for skin cancer sites with an outlook on lipid-based delivery vehicles and diagnostic imaging inputs for skin conditions

Skin cancer is among one of the most common human malignancies wide-spread world-over with mortality statistics rising continuously at an alarming rate. The increasing frequency of these malignancies has marked the need for adopting effective treatment plan coupled with better and site-specific delivery options for the desired therapeutic agent's availability at the affected site. The concurrent delivery approaches to cancerous tissues are under constant challenge and, as a result, are evolving and gaining advancements in terms of delivery modes, therapeutic agents and site-specificity of the therapeutics delivery. The lipid-based liposomal drug delivery is an attractive and emerging option, and which is meticulously shaping up beyond a threshold level to a promising, and viable route for the effective delivery of therapeutic agents and other required injuctions to the skin cancer. An update on liposomal delivery of chemotherapeutic agents, natural-origin compounds, photosensitizer, and DNA repair enzymes as well as other desirable and typical delivery modes employed in drug delivery and in the treatment of skin cancers is discussed in details. Moreover, liposomal delivery of nucleic acid-based therapeutics, i.e., small interfering RNA (siRNA), mRNA therapy, and RGD-linked liposomes are among the other promising novel technology under constant development. The current clinical applicability, viable clinical plans, future prospects including transport feasibility of delivery vesicles and imaging techniques in conjunction with the therapeutic agents is also discussed. The ongoing innovations in liposomal drug delivery technology for skin cancers hold promise for further development of the methodology for better, more effective and site-specific delivery as part of the better treatment plan by ensuring faster drug transport, better and full payload delivery with enough and required concentration of the dose. Summary: The study isolated phenanthroindolizidine alkaloids from Tylophora indica and evaluated their anticancer activity. Among the isolated compounds, tylophorinidine (5) exhibited the highest cytotoxicity against three different cancer cell lines. Molecular docking studies confirmed the bioactivity of these compounds.

Summary: The study evaluates a newly modified cyclodextrin derivative, water-soluble beta-cyclodextrin-epichlorohydrin (beta-CD), as a drug carrier to improve the solubility and bioavailability of galangin (GAL). In vitro tests showed that the GAL/beta-CD complex exhibited cytotoxic effects on cancer cells without affecting normal cells. In vivo experiments on mice demonstrated the lack of toxic effects on normal cells.

SPIONs Conjugate Supported Anticancer Drug Doxorubicin's Delivery: Current Status, Challenges, and Prospects

Summary: Considerable efforts have been made to develop nano-structured carriers, based on superparamagnetic iron oxide nanoparticles (SPIONs), for the delivery of the anticancer drug doxorubicin. These carriers have shown promise in overcoming the limitations of doxorubicin's toxicity, low therapeutic outcomes, inappropriate dosing, metastatic and general resistance, and non-selectivity. The development of SPIONs-based drug delivery modules allows for targeted and stable delivery of doxorubicin to different cancer sites, making it a viable option for cancer treatment.

Ciprofloxacin-Loaded Silver Nanoparticles as Potent Nano-Antibiotics against Resistant Pathogenic Bacteria

Summary: In this study, silver nanoparticles (AgNPs) with antibacterial properties were successfully synthesized using the chemical reduction method, and the antibiotic ciprofloxacin (CIP) was loaded onto the AgNPs to enhance its antibacterial activity. The new nanocomposite, AgNPs-PEG-CIP, showed higher antibacterial, antibiofilm, and antioxidant activity against the tested pathogenic bacteria compared to CIP alone, indicating its potential as an antibacterial agent.

Combined Anti-Bacterial Actions of Lincomycin and Freshly Prepared Silver Nanoparticles: Overcoming the Resistance to Antibiotics and Enhancement of the Bioactivity

Summary: Bacterial drug resistance to antibiotics is increasing globally, and scientists are continuously developing strategies to overcome this issue. In this study, the efficacy of combining silver nanoparticles and lincomycin antibiotic in combatting bacterial resistance was demonstrated. The combined delivery showed improved antibacterial activity compared to the individual use of silver nanoparticles and lincomycin in Bacillus cereus and Proteus mirabilis microorganisms. Additionally, the study found that silver nanoparticles exhibit strong antioxidant activity.

Summary: This study aimed to develop a cosmeceutical preparation using the strong antioxidant potential of Ginkgo biloba (GB) leaves. Cream incorporated GB (GBC) was prepared and characterized, showing stable physical and chemical properties with a pH similar to the skin. The cream showed effectiveness and safety, with antioxidant and rejuvenating effects on the skin during the two-week trial.

Summary: Halophytes and xerophytes, plants with strong tolerance to salinity and drought, have higher production of phenolics and flavonoids compared to normal vegetation. Due to increasing desertification and the associated challenges of high salinity, temperature, and water scarcity, these plants have become important for environmental protection, land reclamation, and food security. They also show potential in the development of newer cancer therapeutics, as they possess prophylactic properties and exhibit anti-cancer effects through their phytochemical and pharmacological properties, particularly immunomodulation.

Phytochemical and biological activity profiles of Thymbra linearifolia: An exclusively native species of Libyan Green mountains

Summary: This article presents the first documented phytochemical and biological evaluation report of Thymbra linearifolia, an endemic Libyan plant. The study revealed the presence of phenolic and flavonoid compounds in the plant, which exhibited strong antioxidant and anticancer activities. Additionally, the plant also showed moderate antimicrobial effects.

Summary: Combining bovine serum albumin with microemulsions as a biopolymer ingredient is an innovative method for addressing surface functionalization and stability issues in targeted payload deliveries. The current study aimed to develop an efficient microemulsion system encapsulating sesame oil as a model payload for efficient delivery. Characterization and analysis of the developed carrier were performed using UV-VIS, FT-IR, and FE-SEM. Physicochemical properties of the microemulsion were assessed and analyzed using dynamic light scattering, zeta potential, electron microscopy, and rheological behavior. In vitro and in vivo assays were conducted to evaluate cell viability, biocompatibility, and toxicity.

Graphene oxide-induced, reactive oxygen species-mediated mitochondrial dysfunctions and apoptosis: high-dose toxicity in normal cells

Summary: In this study, the cytotoxic effects of graphene oxide nanoparticles (GONPs) were evaluated using MTT assays, observation of apoptotic markers, and oxidative stress. Significant cytotoxic and apoptotic effects were observed. Analyses of CYP2E1 and malondialdehyde concentrations confirmed the toxicity of GONPs, while the levels of glutathione showed a reduction compared to the control. The alterations in cytochrome CYP2E1, glutathione, malondialdehyde, and caspase-3 suggest an interlinked relationship.

Variability in the volatile constituents and biological activities of Achillea millefolium L. essential oils obtained from different plant parts and by different solvents

Summary: Achillea millefolium Linn. is traditionally used for its medicinal properties and has economic value in cosmetic and insect repellent industries. The essential oils obtained from different parts of the plant were analyzed and investigated for their antioxidant and anticancer activities. The major constituents of the volatile oils varied among different batches of the plant, with monoterpenes being predominant in the distilled oils and sesquiterpenes in the non-polar extracts. The distilled batches exhibited better antioxidant activity, while the non-polar extracts showed remarkable cytotoxic effects against cancer cells.

Effects of DHA on cognitive dysfunction in aging and Alzheimer's disease: The mediating roles of ApoE

Summary: The prevalence of Alzheimer's disease is increasing due to aging population, and ApoE4 allele is identified as the primary genetic risk factor. However, emerging research suggests that dietary factors, such as DHA, can modify the risk of AD. The interplay between DHA and ApoE genotype within the brain needs further investigation. Early DHA supplementation may benefit cognitively normal older adults carrying the ApoE4 gene, while its effectiveness for AD patients is limited.

Advances in research on microbial conjugated linoleic acid bioconversion

The review aims to serve as a reference for microbial CLA production and expand the understanding of the potential probiotic role of microbial CLA producers.
