C2-substituted aromatic cytokinin sugar conjugates delay the onset of senescence by maintaining the activity of the photosynthetic apparatus

Cytokinins are plant hormones with biological functions ranging from coordination of plant growth and development to the regulation of senescence. A series of 2-chloro-N-6-(halogenobenzylamino)purine ribosides was prepared and tested for cytokinin activity in detached wheat leaf senescence, tobacco callus and Amaranthus bioassays. The synthetic compounds showed significant activity, especially in delaying senescence in detached wheat leaves. They were also tested in bacterial receptor bioassays using both monocot and dicot members of the cytokinin receptor family. Most of the derivatives did not trigger cytokinin signaling via the AHK3 and AHK4 receptors from Arabidopsis thaliana in the bacterial assay, but some of them specifically activated the ZmHK1 receptor from Zea mays and were also more active than the aromatic cytokinin BAP in an ARR5::GUS cytokinin bioassay using transgenic Arabidopsis plants. Whole transcript expression analysis was performed using an Arabidopsis model to gather information about the reprogramming of gene transcription when senescent leaves were treated with selected C2-substituted aromatic cytokinin ribosides. Genome-wide expression profiling revealed that the synthetic halogenated derivatives induced the expression of genes related to cytokinin signaling and metabolism. They also prompted both up- and down -regulation of a unique combination of genes coding for components of the photosystem II (PSII) reaction center, light -harvesting complex II (LHCII), and the oxygen -evolving complex, as well as several stress factors responsible for regulating photosynthesis and chlorophyll degradation. Chlorophyll content and fluorescence analyses demonstrated that treatment with the halogenated derivatives increased the efficiency of PSII photochemistry and the abundance of LHCII relative to DMSO- and BAP -treated controls. These findings demonstrate that it is possible to manipulate and fine-tune leaf longevity using synthetic aromatic cytokinin analogs. (C) 2015 Elsevier Ltd. All rights reserved.

Summary: This study demonstrates how multivariate statistical analysis tools can be used to simplify omics data and extract relevant information in order to further understand the use of biostimulants in promoting plant growth and alleviating stress. The study found that lettuce and tomato plants exhibited different phenotypic and metabolomic responses to biostimulants, with biomass-related parameters being the most relevant traits to evaluate the biostimulant mode of action in lettuce, while chlorophyll fluorescence parameters and certain antistress metabolites were the relevant traits in tomatoes.

Cytokinin oxidase/dehydrogenase inhibitors: outlook for selectivity and high efficiency

Seed germination and in vitro propagation of three threatened endemic South African Aloe species

Summary: In this study, four new cytokinin salts were synthesized and found to have higher water solubility compared to traditional cytokinins. Foliar treatment with these salts significantly reduced chlorophyll degradation and impairment of photosystem II functionality in barley leaf segments undergoing artificial senescence and maintained high concentrations of functional metabolites for several days.

Presence and future of plant phenotyping approaches in biostimulant research and development

High-throughput plant phenotyping can accelerate the development and characterization of new biostimulants.

Carbon dioxide expanded liquid: an effective solvent for the extraction of quercetin from South African medicinal plants

Summary: The study successfully extracted quercetin from quince fruit using an environmentally friendly method, with optimized conditions of a CO2/ethanol/water ratio of 10/81/9, extraction temperature of 66 degrees Celsius, pressure of 22.3 MPa, and extraction time of 30 minutes. The method showed good repeatability and was applicable to various plant samples.

Topolin cytokinins enhanced shoot proliferation, reduced hyperhydricity and altered cytokinin metabolism in Pistacia vera L. seedling explants

Summary: The effects of mT and mTR on the proliferation and anomalies of Pistacia vera L. were studied and compared to that of BA. The results showed that mT had the highest proliferation rate and mTR had the lowest percentage of hyperhydric usable shoots and callus weight. However, leaf necrosis and shoot tip were not influenced by the different cytokinins. Image analysis revealed that BA and mTR had higher photosynthetic efficiency, but BA-treated shoots had higher anthocyanin accumulation, suggesting more stress. The study also investigated the metabolism of exogenous BA, mT, and mTR in pistachio.

The Role of a Cytokinin Antagonist in the Progression of Clubroot Disease

Study on late-stage infected Arabidopsis thaliana showed changes in cytokinin levels and their role in disease progression.

Rare 7,9′-dinorlignans with neuroprotective activity from the roots of Lindera aggregata (Sims) Kosterm
