Summary: This study evaluated the therapeutic efficacy of a novel PI3K inhibitor, DHW-208, in hepatocellular carcinoma (HCC) and investigated its underlying mechanism. The results showed that DHW-208 could effectively inhibit the proliferation, migration, invasion, and angiogenesis of HCC through the PI3K/AKT/mTOR signaling pathway in vitro. In vivo studies further confirmed the antitumor effect of DHW-208 by inhibiting the PI3K/AKT/mTOR pathway with a high degree of safety. These findings suggest that DHW-208 has potential as a small molecule PI3K inhibitor for HCC treatment and provide a theoretical basis for the development of PI3K inhibitors.

Summary: This study identified a novel circRNA ASPH (circASPH) that mediates hepatic fibrosis (HF) by targeting the miR-139-5p/Notch1 axis. The expression profile of circASPH in hepatocyte exosomes of mice with HF was found to be upregulated. Downregulation of circASPH expression inhibited hepatic stellate cell (HSC) activation and proliferation, induced apoptosis, and attenuated mouse liver fibrogenic injury. Thus, circASPH may serve as a potential biomarker for HF diagnosis and therapy.

Summary: This study investigated the specific mechanism by which TROAP functions in kidney renal clear cell carcinoma (KIRC). It was found that TROAP was significantly overexpressed in KIRC tissues and was associated with higher T and pathological stages, and a poor prognosis. Inhibition of TROAP expression reduced KIRC proliferation, affected cell cycle, promoted cell apoptosis, and reduced cell migration and invasion. Further analysis revealed that TROAP may regulate KIRC progression by binding to STAT3.

Summary: In this study, we found that high expression of ARF6 is associated with poor prognosis and promotes the proliferation of hepatocellular carcinoma (HCC). ARF6 was upregulated in HCC tissues and correlated with poor tumor differentiation, incomplete tumor encapsulation, advanced tumor TNM stage, and poor prognosis. ARF6 activated the STAT3 signaling pathway to enhance HCC cell proliferation and tumor growth. These findings suggest that ARF6 may serve as a potential prognostic and therapeutic target for HCC patients.
