Natural products have always been sought as a dependable source for the cure of many fatal diseases including cancer. Resveratrol (RSV), a naturally occurring plant polyphenol, has been of recent research interest and is being investigated for its beneficial biological properties that include antioxidant, anti-inflammatory, proapoptotic, and growth inhibitory activities. These effects are mainly mediated by cell cycle arrest, upregulation of proapoptotic proteins, loss of mitochondrial potential, and generation of reactive oxygen species. Among the beneficial properties of RSV, the anticancer property has been of the prime focus and extensively explored during the last few years. Although reports exist on the chemopreventive role of RSV in many solid tumors, limited information is available on the antiproliferative activity of RSV in human lymphoma cells and experimental models. Potential mechanisms for its antiproliferative effect include induction of cell differentiation, apoptosis, and inhibition of DNA synthesis. In this review, the different kinds of lymphoid malignancies and the main mechanisms of cell death induced by resveratrol are discussed. The challenges are limiting in vivo experimental studies involving resveratrol. An attempt for the translation of this compound into a clinical drug also forms a part of this review.

Activation of NOTCH signaling via DLL1 is mediated by APE1-redox-dependent NF-κB activation in oesophageal adenocarcinoma

Lei Chen, Heng Lu, Dunfa Peng, Long Long Cao, Farah Ballout, Kannappan Srirmajayam, Zheng Chen, Ajaz Bhat, Timothy C. Wang, Anthony Capobianco, Jianwen Que, Oliver Gene McDonald, Alexander Zaika, Shutian Zhang, Wael El-Rifai

Summary: This study investigates the role of acidic bile salts in activating NOTCH signaling in oesophageal adenocarcinoma (EAC). The researchers found that there is a crosstalk between APE1 redox function, reflux-induced inflammation, and DLL1 upregulation, which promotes cancer cell stem-like properties in EAC. The overexpression of APE1 and DLL1 is associated with poor overall survival in EAC patients.

Summary: Long non-coding RNAs (lncRNAs) play crucial roles in cancer tumorigenesis and progression. Recent studies have shown that nuclear paraspeckle assembly transcript 1 (NEAT1) as an lncRNA can regulate cell proliferation, migration, and EMT in tumor cells. NEAT1 regulates cancer cell behavior by targeting several miRNA/mRNA axes. Therefore, NEAT1 may serve as a potential biomarker for predicting and treating certain human cancers. In this review, we provide an overview of various NEAT1-related signaling pathways that are critical in cancer initiation and progression.

Expression profile of KIR3DS1/KIR3DL1 receptors in association with immunological responses in TB, HIV and HIV/TB infected patients

Sobia Naz Shaukat, Faizan Nasir, Afsheen Raza, Rafiq Khanani, Shahab Uddin, Shahana Urooj Kazmi

Summary: This study aimed to evaluate the association between KIR3DS1 and KIR3DL1 with IFN-y/IL-10 in HIV-1 and TB infections. The results showed that expression of KIR3DS1 was associated with low HIV-1 viral load, while expression of KIR3DL1 was significantly upregulated in HIV-1. In addition, IFNy expression was decreased in TB but increased in HIV-1/TB, while IL-10 expression was increased in HIV-1 and HIV-1/TB. It suggests that KIR3DS1/L1 expression is associated with IFNy/IL-10 responses and modulates disease severity in HIV-1 and TB infections.

Investigating the Use of Machine Learning Models to Understand the Drugs Permeability Across Placenta

Vaisali Chandrasekar, Mohammed Yusuf Ansari, Ajay Vikram Singh, Shahab Uddin, Kirthi S. Prabhu, Sagnika Dash, Souhaila Al Khodor, Annalisa Terranegra, Matteo Avella, Sarada Prasad Dakua

Summary: Due to limited drug testing options for pregnant women, the development of computational algorithms is vital in predicting drug behavior in the placental barrier as an alternative to animal testing. The ability of a molecule to cross the placental barrier and affect the fetus is crucial in determining its toxicological effects. Through analyzing publicly available datasets, this study explores different machine learning models, fingerprints, and toolkits to find the most suitable models. Neural network-based models show effective predictions of permeability. Among them, K-nearest neighbor (KNN), standard vector classifier (SVC), and Multi-layer perceptron (MLP) perform the best with prediction percentages of 82%, 86.4%, and 90.8% respectively. Comparison of different models reveals negative permeability predictions for drugs like Aliskiren, certain insulin secretagogues, and glucocorticoids.
