A novel lateral flow assay based on GoldMag nanoparticles and its clinical applications for genotyping of MTHFR C677T polymorphisms

Current techniques for single nucleotide polymorphism (SNP) detection require tedious experimental procedures and expensive and sophisticated instruments. In this study, a visual genotyping method has been successfully established via combining ARMS-PCR with gold magnetic nanoparticle (GoldMag)based lateral flow assay (LFA) and applied to the genotyping of methylenetetrahydrofolate reductase (MTHFR) C677T. C677T substitution of the gene MTHFR leads to an increased risk of diseases. The genotyping result is easily achievable by visual observation within 5 minutes after loading of the PCR products onto the LFA device. The system is able to accurately assess a broad detection range of initial starting genomic DNA amounts from 5 ng to 1200 ng per test sample. The limit of detection reaches 5 ng. Furthermore, our PCR-LFA system was applied to clinical trials for screening 1721 individuals for the C677T genotypes. The concordance rate of the genotyping results detected by PCR-LFA was up to 99.6% when compared with the sequencing results. Collectively, our PCR-LFA has been proven to be rapid, accurate, sensitive, and inexpensive. This new method is highly applicable for C677T SNP screening in laboratories and clinical practices. More promisingly, it could also be extended to the detection of SNPs of other genes.

A Special Case of Relapsing-Remitting Bilateral Encephalitis: Without Epilepsy, but Responding to Rituximab and with a Brain Biopsy Coinciding with Rasmussen Encephalitis

Summary: A nine-year-old boy presented with headache, cognitive decline, and hemiparesis. Brain MRI revealed bilateral cortical lesions with new lesions developing over five years. Initial diagnosis of primary angiitis of the central nervous system was considered. Treatment with corticosteroids, immunoglobulins, and cyclophosphamide worsened symptoms. Brain biopsy after five years showed pathology changes consistent with RE. Treatment with rituximab resulted in stable lesions on follow-up MRI. This case highlights the uniqueness and treatment challenges of this immune-mediated condition.

ZDHHC16 restrains osteogenic differentiation of bone marrow mesenchymal stem cells by inhibiting phosphorylation of CREB

In a mouse femur fracture model, a general inhibitor of ZDHHCs, 2-BP, was found to promote fracture healing.

Chronic salmon calcitonin exerts an antidepressant effect via modulating the p38 MAPK signaling pathway

Summary: Depression is a prevalent mental disorder that has a high suicide rate. Current medications for depression have limitations, thus there is a need for more efficient and safe drugs. This study investigated the potential of salmon calcitonin (sCT) as an antidepressant and identified the molecular mechanism behind its effects. Results showed that sCT downregulated the phosphorylation of p38/JNK kinases in the hippocampus of mice exposed to chronic stress, suggesting its potential as a treatment for depression.

It is validated to achieve desirable performance in interpreting genetic profiles for drug responses and can be reprogrammed for other pharmacogenetics genes and risk assessments for personalized healthcare.

Screening a DNA Aptamer Specifically Targeting Integrin & beta;3 and Partially Inhibiting Tumor Cell Migration

Summary: Due to its key roles in malignant tumor progression and reprogramming of the tumor microenvironment, integrin β3 has attracted great attention as a new target for tumor therapy. This study used a purified extracellular domain of integrin β3 and integrin β3-positive cells to screen aptamers specifically targeting integrin β3 in its native conformation on live cells. The optimized aptamer S10yh2 showed high binding affinity and selectivity, low equilibrium dissociation constant, and the ability to downregulate integrin αvβ3 levels and partially inhibit cell migration behavior.

Metal oxide-embedded carbon-based materials for polymer solar cells and X-ray detectors

Summary: This study examines the effects of hybrid nanoparticles made of NiO@rGO and NiO@CNT on the active layers of polymer solar cells and X-ray photodetectors. Among the tested configurations, the NiO@CNT device demonstrates superior performance in converting sunlight into electricity, and achieves the best sensitivity for X-ray detection.

In situ growth of a redox-active metal-organic framework on electrospun carbon nanofibers as a free-standing electrode for flexible energy storage devices

Summary: The rational construction of free-standing and flexible electrodes for electrochemical energy storage devices is an emerging research focus. In this study, a redox-active metal-organic framework (MOF) was prepared on carbon nanofibers using an in situ approach, resulting in a flexible electrode with high redox-active behavior and unique properties such as high flexibility and lightweight. The prepared electrode showed excellent cyclic retention and rate capability in supercapacitor applications. Additionally, it could be used as a freestanding electrode in flexible devices at different bending angles.

A NIR-driven green affording-oxygen microrobot for targeted photodynamic therapy of tumors

Summary: Photodynamic therapy (PDT) is a promising local treatment modality in cancer therapy, but its therapeutic efficacy is restricted by ineffective delivery of photosensitizers and tumor hypoxia. In this study, a phototactic Chlorella-based near-infrared (NIR) driven green affording-oxygen microrobot system was developed for enhanced PDT. The system exhibited desirable phototaxis and continuous oxygen generation, leading to the inhibition of tumor growth in mice. This study demonstrates the potential of using a light-driven green affording-oxygen microrobot to enhance photodynamic therapy.

Novel hollow MoS2@C@Cu2S heterostructures for high zinc storage performance

Facile green synthesis of wasted hop-based zinc oxide nanozymes as peroxidase-like catalysts for colorimetric analysis

The WHE-ZnO NZs exhibit exceptional peroxidase-like activity and serve as effective catalysts for the oxidation of 3,3,5,5-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide (H2O2). In addition, a straightforward colorimetric technique for detecting both H2O2 and glucose was developed using the WHE-ZnO NZs as peroxidase-like catalysts.

Impact of channel nanostructures of porous carbon particles on their catalytic performance

Summary: Mesoporous carbon particles have unique structural properties that make them suitable as support materials for catalytic applications. This study investigates the impact of channel nanostructures on the catalytic activity of porous carbon particles (PCPs) by fabricating PCPs with controlled channel exposure on the carbon surface. The results show that PCPs with highly open channel nanostructures exhibit significantly higher catalytic activity compared to those with closed channel nanostructures.

Strongly coupled plasmonic metal nanoparticles with reversible pH-responsiveness and highly reproducible SERS in solution

The pH-responsive polymer-grafted nanoparticles show high reproducibility and sensitivity in solution, providing a novel approach for SERS without the need for sample pre-concentration.
