Summary: Research findings suggest that overexpression of mortalin in breast cancer is associated with poorer overall survival rates, with its activation of the Wnt/GSK3β/β-catenin signaling pathway promoting Epithelial-mesenchymal transition (EMT) and maintaining the stemness of breast cancer stem cells (BCSCs).

Summary: PPP1R14C is upregulated in TNBC and associated with poor prognosis. Overexpression of PPP1R14C promotes cell proliferation and the aggressive phenotype in TNBC cells, while depletion of PPP1R14C has the opposite effect. Additionally, phosphorylated PPP1R14C inhibits the phosphorylation of GSK3β and recruits E3 ligase TRIM25 for ubiquitylation and degradation of non-phosphorylated GSK3β.

Summary: Mitomycin treatment induces pulmonary toxicity by causing alveolar epithelial cell senescence, with GSK3 beta (S9) phosphorylation playing a crucial role in this process. Inhibition of Akt1 and knockdown of p53 can suppress the senescence of alveolar epithelial cells induced by mitomycin. Baicalein treatment weakens GSK3 beta (S9) phosphorylation and alleviates mitomycin-induced senescence of alveolar epithelial cells. Targeting GSK3 beta (S9) phosphorylation may offer a potential approach to attenuate mitomycin-induced pulmonary toxicity.

Summary: In this study, the anti-epithelial-mesenchymal transition (EMT) and antifibrotic effects of Honokiol in liver fibrosis were explored. It was found that Honokiol can suppress TGF-beta 1-induced EMT and migration of liver cells and inhibit hepatic fibrosis by regulating the E-cadherin/GSK3 beta/JNK pathway and inhibiting the AKT/ERK/p38/beta-catenin/TMPRSS4 signaling axis.

Summary: Research has shown that baicalein (BAI) can inhibit gastric cancer cell growth by suppressing cell proliferation, migration, and inducing apoptosis, as well as downregulating the AKT/mTOR signaling pathway. BAI may be a potential therapeutic factor for treating gastric cancer.

Summary: PRDX6, a member of the peroxiredoxin family, exhibits peroxidase, phospholipase A2 (PLA2), and lysophosphatidylcholine (LPC) acyltransferase (LPCAT) activities. Knockout of PRDX6 in SNU475 hepatocarcinoma cells leads to lipid peroxidation, mitochondrial dysfunction, metabolic reprogramming, altered cytoskeleton, and down-regulation of growth-related proteins. Activation of upstream regulators and inhibition of migration and invasiveness suggest the involvement of PRDX6 in tumor development and metastasis.

Summary: This study demonstrates the synergistic anti-cancer effect of the combination of lidocaine and palbociclib on breast cancer cells by inhibiting the PI3K/AKT/GSK3 beta pathway. The combination treatment significantly inhibits tumor growth and induces apoptosis, suggesting that it could be an effective therapy for breast cancer.

Summary: This study investigates the molecular docking and dynamics simulations of alpha-mangostin on glycogen synthase kinase 3 beta (GSK3 beta). The results show that alpha-mangostin has a weaker binding affinity to GSK3 beta compared to the native ligand but shares similar docking sites. This suggests that alpha-mangostin may have potential as an anti-TNBC agent.

Summary: The compound MC0704, a synthetic STAT3 pathway inhibitor, showed potential antitumor activity in docetaxel-resistant TNBC cells. It effectively inhibited the metastatic potential of the cells in vitro and exhibited strong antitumor activity when combined with docetaxel in a xenograft mouse model. MC0704 could be a potential therapeutic agent for TNBC patients with docetaxel resistance.
