Summary: This study investigates the relationship between B7-H4 and immune evasion, cancer stemness, and epithelial-mesenchymal transition in ovarian serous carcinoma. The results show that B7-H4 expression is positively correlated with the degree of tumor-infiltrating lymphocytes, as well as the expression of CD24, CD44s, CD133, and E-cadherin. B7-H4 expression is inversely correlated with the expression of ALDH1, N-cadherin, and vimentin. In addition, B7-H4 positive patients have higher tumor grade and lower overall survival rate, especially in cases with low stromal TILs.

Summary: This study developed immune scoring systems focusing on B7-H3, B7-H4, and HHLA2, and demonstrated their potential in predicting survival and immunotherapeutic efficacy for gallbladder cancer (GBC). The results showed that these immune scoring systems were associated with worse clinicopathological stage and unfavorable prognosis in postoperative survival. Additionally, novel prediction models based on immune stratification and TNM staging showed excellent discrimination ability in predicting survival and immunotherapeutic efficacy for GBC patients.

Summary: Antibody-drug conjugates (ADC) achieve targeted drug delivery to tumors and have shown clinical success. The construction of an ADC, including antibody, payload, linker, and conjugation method, as well as drug-to-antibody ratio (DAR), affects its activity and safety profile. A novel ADC platform called Dolasynthen (DS) based on auristatin hydroxypropylamide payload enables precise DAR-ranging and site-specific conjugation. XMT-1660, a site-specific DS DAR 6 ADC targeting B7-H4, exhibited complete tumor regressions in breast and ovarian cancer models and showed activity in a breast cancer model refractory to PD-1 immune checkpoint inhibition, indicating its potential as a therapeutic option.
