Summary: The process of mammary development in humans reveals a connection between mammary cells and skin epithelial cells. Some breast tumors were found to express involucrin, a keratinocyte differentiation marker, suggesting a potential de-differentiation to a keratinocyte-derived state. This hypothesis was confirmed by studying the frequency of involucrin expression along with Brk, a tyrosine kinase that is expressed in breast carcinomas and differentiating epithelial cells. Interestingly, breast carcinoma samples with high involucrin levels also expressed Brk. Further research showed that 1,25-dihydroxyvitamin D3, a differentiation and potential anti-cancer agent, could decrease proliferation in breast cancer cell lines expressing both involucrin and Brk. These findings contribute to our understanding of cellular responses to 1,25-dihydroxyvitamin D3 independent of vitamin D receptor status and provide potential markers for predicting therapeutic responses to this agent.

Summary: Noncompliance in longitudinal RCTs can complicate treatment effect assessment. The complier average causal effect (CACE) has been used to evaluate intervention efficacy for compliant subpopulations. Traditional univariate analysis may be limited in power to detect CACE due to small sample sizes, low compliance rates, and individual endpoint effect sizes. To overcome this, a multivariate longitudinal potential outcome model is proposed to efficiently assess multivariate CACE by combining information across multiple endpoints and visits.
