This article provides a brief description of the anticancer potential of natural compounds, followed by a search for patents published from 2019 onwards according to the PRISMA guidelines. Additionally, relevant publications from the overall scientific literature were discussed.
Introduction: Exploring the chemical diversity and molecular mechanisms of natural products continues to be an important research area for identifying novel promising therapeutic approaches for fighting cancer. This is a complex disease and poses important challenges, which require not only targeted interventions to improve chemotherapy efficacy and tolerability, but also adjuvant strategies to counteract chemoresistance development and relapses.Areas covered: After a brief description of the recent literature on the anticancer potential of natural compounds, we searched for patents following the PRISMA guidelines, filtering the results published from 2019 onwards. In addition, some relevant publications from the overall scientific literature were also discussed.Expert opinion: This review comprehensively covers and analyzes the most recent advances on the anticancer mechanism of licensed natural compounds and their chemical optimization. Patentability of natural compounds was discussed according to the recent legislation in the U.S.A. and Europe. [GRAPHICAL ABSTRACT]

Coumarin-pyrazoline Hybrids as Selective Inhibitors of the Tumor-associated Carbonic Anhydrase IX and XII

Summary: This study synthesized a series of coumarin derivatives incorporating pyrazole-1-carboxamide moiety and evaluated their inhibitory activity against carbonic anhydrase. The results showed that these compounds did not inhibit the widespread CA I and II, but showed sub-micromolar inhibitory activity against CA IX and XII with high selectivity.

Discovery of Chalcone-Based Hybrid Structures as High Affinity and Site-Specific Inhibitors against SARS-CoV-2: A Comprehensive Structural Analysis Based on Various Host-Based and Viral Targets

Summary: Previous studies have shown that natural-based chalcones can effectively inhibit coronavirus enzymes and modulate antiviral targets. This study investigated the inhibitory effects of 757 chalcone-based compounds on the enzymes and selected host-based targets. The results identified CHA-12 as the most potent multi-target inhibitor, and specific fragments in certain compounds were found to be effective in inhibiting the enzymes. The discovery of CHA-12 as a potential treatment for both respiratory symptoms and COVID-19 infection is significant.

Antibiofilm activity and NMR-based metabolomic characterization of cell-free supernatant of Limosilactobacillus reuteri DSM 17938

Summary: This study evaluated the antibiofilm activity of cell-free supernatant (CFS) and its sub-fractions (SurE 10 K and SurE) produced by Limosilactobacillus reuteri DSM 17938 against biofilm-producing bacterial species. The results showed that CFS exhibited promising antibiofilm activity against biofilms formed by clinically relevant microorganisms. NMR metabolomic analysis identified and quantified several compounds, mainly organic acids and amino acids, with lactate being the most abundant metabolite in all samples. The storage stability of these postbiotics was evaluated using the CIEL*a*b parameters.

Nrf2 activation may play a role in these properties. Further research can increase interest in this product.

Summary: Tylophorine and lycorine, two potential antiviral agents derived from plants, show broad-spectrum antiviral properties and anti-inflammatory effects. They may help combat respiratory viral infections, mitigate the oncogenic effects of viruses, and contribute to cancer research. However, further research is needed to address pharmacologic development issues.

Novel Insights on CAI-CORM Hybrids: Evaluation of the CO Releasing Properties and Pain-Relieving Activity of Differently Substituted Coumarins for the Treatment of Rheumatoid Arthritis

Summary: Pain control is crucial in rheumatoid arthritis (RA) patients, but current treatments have limited effectiveness and risk of side effects. In this study, we proposed a novel approach combining carbonic anhydrase inhibitors (CAIs) with CO releasing molecules (CORMs). The resulting CAI-CORM hybrids showed significant anti-inflammatory effects and pain relief in RA rat models. Our findings demonstrate the potential of CAI-CORM hybrids as anti-nociceptive agents against arthritis.

Characterization of the Chemopreventive Properties of Cannabis sativa L. Inflorescences from Monoecious Cultivars Grown in Central Italy

Summary: Hemp bioproducts, with their diverse bioactive compounds and potential health benefits, show great promise for nutraceutical and pharmaceutical applications. This study investigated the chemically characterized hydroalcoholic and organic extracts obtained from locally cultivated hemp varieties and found that they possess antimutagenic, radical scavenging, and cytotoxic properties. These findings suggest that bioactive compounds in hemp inflorescences could be harnessed for genoprotective and chemopreventive purposes.
