ACSL1-induced ferroptosis and platinum resistance in ovarian cancer by increasing FSP1 N-myristylation and stability

Reprogramming of lipid metabolism affects energy utilization and cell signaling in cancer cells, contributing to cell survival and metastasis. Ferroptosis, a form of necrosis caused by lipid oxidation overload, has been implicated in cancer metastasis. However, the mechanism by which fatty acid metabolism regulates anti-ferroptosis signaling pathways is not fully understood. This study demonstrates that the formation of ovarian cancer spheroids and exposure to platinum chemotherapy increase levels of anti-ferroptosis proteins, including ACSL1. Inhibition of ferroptosis enhances spheroid formation, while genetic manipulation of ACSL1 expression reduces lipid oxidation and increases resistance to ferroptosis by modulating the myristoylation of FSP1.
Reprogramming of lipid metabolism, which modulates energy utilization and cell signaling, maintains cell survival and promotes cancer metastasis in cancer cells. Ferroptosis is a type of cell necrosis caused by an overload of lipid oxidation, which has been demonstrated to be involved in cancer cell metastasis. However, the mechanism by which fatty acid metabolism regulates the anti-ferroptosis signaling pathways is not fully understood. The formation of ovarian cancer spheroids helps to counteract the hostile microenvironment of the peritoneal cavity with low oxygen, shortage of nutrients, and subjected to platinum therapy. Previously, we demonstrated that Acyl-CoA synthetase long-chain family member 1 (ACSL1) promotes cell survival and peritoneal metastases in ovarian cancer, but the mechanism is still not well elucidated. In this study, we demonstrate that the formation of spheroids and under exposure to platinum chemotherapy increased the levels of anti-ferroptosis proteins as well as ACSL1. Inhibition of ferroptosis can enhance spheroid formation and vice versa. Genetic manipulation of ACSL1 expression showed that ACSL1 reduced the level of lipid oxidation and increased the resistance to cell ferroptosis. Mechanistically, ACSL1 increased the N-myristoylation of ferroptosis suppressor 1 (FSP1), resulting in the inhibition of its degradation and translocation to the cell membrane. The increase in myristoylated FSP1 functionally counteracted oxidative stress-induced cell ferroptosis. Clinical data also suggested that ACSL1 protein was positively correlated with FSP1 and negatively correlated with the ferroptosis markers 4-HNE and PTGS2. In conclusion, this study demonstrated that ACSL1 enhances antioxidant capacity and increases ferroptosis resistance by modulating the myristoylation of FSP1.

Summary: This paper presents an intelligent formation operation mode (IFOM) based on straddle-type rapid transit vehicles. The operation mode of the vehicle in the straddle-type rapid transit system is defined in the control system architecture. An evaluation index system for formation vehicles is proposed considering the operating environment. A formation vehicle operation controller is built based on the vehicle longitudinal dynamics model and the artificial potential field formation algorithm. A multi-rigid body dynamics simulation model of formation vehicles in a heterogeneous operating environment is established. The operating performance of formation vehicles in a heterogeneous operating environment is analyzed.

COMMD3-Mediated Endosomal Trafficking of HER2 Inhibits the Progression of Ovarian Carcinoma

Summary: In this study, COMMD3 was identified as a key regulator in the endosomal trafficking of HER2, and its decreased expression was found to inhibit the proliferation, migration, and epithelial-mesenchymal transition of ovarian carcinoma cells. COMMD3 also suppressed tumor growth, formation of malignant ascites, and peritoneal dissemination of cancer cells. Further analysis revealed that COMMD3 interacts with HER2 to direct it to the slow recycling pathway and attenuate downstream tumor-promoting signaling.

From PROTAC to inhibitor: Structure-guided discovery of potent and orally bioavailable BET inhibitors

Summary: An X-ray structure of a CLICK chemistry-based BET PROTAC bound to BRD2(BD2) inspired the synthesis of JQ1 derived heterocyclic amides. These compounds displayed improved profiles compared to JQ1 and birabresib as potent BET inhibitors. A specific compound 1q (SJ1461) showed excellent affinity to both BRD4 and BRD2 and high potency in acute leukaemia and medulloblastoma cell lines. The co-crystal structure of 1q with BRD4-BD1 revealed polar interactions, explaining the observed affinity improvements. Furthermore, pharmacokinetic studies suggested that the heterocyclic amide moiety improved drug-like features. Our study led to the discovery of the potent and orally bioavailable BET inhibitor 1q (SJ1461) as a promising candidate for further development.

In this study, a novel recombinant human microplasminogen (rh mu Plg) was developed and shown to have potential value in the treatment of vitreoretinopathy.

Integrated transcriptomic analysis identifies coordinated responses to nitrogen and phosphate deficiency in rice

Summary: Nitrogen (N) and phosphorus (P) are essential components of fertilizers for crop production. However, the integration of N and P signaling pathways in plants remains poorly understood. In this study, we investigated the gene expression profiles and physiological responses of rice (Oryza sativa) to N and P deficiency. We found that N and P shortage significantly inhibited rice growth and nutrient uptake. Through the analysis of differentially expressed genes (DEGs), we identified specific physiological reactions and processes induced by N and Pi deficiency. We also discovered a transcriptional regulatory network between N and P signaling pathways, with a focus on the transcription factor gene NITRATE-INDUCIBLE, GARP-TYPE TRANSCRIPTIONAL REPRESSOR 1 (NIGT1), which plays a role in P homeostasis and N acquisition in rice.

Summary: Using the BESIII detector, a sample of (10.09 +/- 0.04) x 109 J/psi decays is analyzed in the (KSKS0)-K-0 pi(0) invariant mass region below 1.6 GeV/c(2). The analyses show dominant pseudoscalar and axial vector components, with good consistency for other individual components. The mass dependent analysis reveals the presence of two isoscalar resonant states, namely the eta(1405) and eta(1475), in the pseudoscalar component.

Summary: This study investigates the alteration of gut microbiota and short-chain fatty acids (SCFAs) in poststroke patients and their relationship with physical condition, intestinal health, pain, and nutritional status. The results demonstrate that poststroke patients have lower community richness of gut microbiota and reduced levels of SCFAs. These changes in gut microbiota are closely associated with physical function, intestinal function, pain, and nutritional status of poststroke patients.

Parity-Time-Symmetric Optoelectronic Oscillator Based on Stimulated Brillouin Scattering

Summary: In this study, a parity-time (PT) symmetric optoelectronic oscillator (OEO) based on stimulated Brillouin scattering (SBS) is proposed and experimentally demonstrated. Instead of using two mutually coupled feedback loops, the PT symmetry is achieved in a single overlapping spatial loop by utilizing SBS gain and SBS loss. Stable single-mode oscillation is achieved by balancing the gain and loss through adjusting the pump power and the polarization states of the probe and pump lights. Experimental results show the generation of a 9.66 GHz microwave signal with a side-mode suppression ratio of 41 dB, and a single-sideband phase noise as low as -120.1 dBc/Hz at a 10 kHz offset frequency. This research opens up new possibilities for enhancing mode selectivity in OEOs and potential integration on a chip.

Summary: This study retrospectively reviewed the characteristics of 788 DLBCL patients and found that 42 patients were diagnosed with multiple primary malignancies (MPMs), and 22 patients were diagnosed with subsequent primary malignancies (SPM). The incidence of SPM was associated with older age, specific subtype, and early clinical stage. MPM was an independent prognostic factor for DLBCL.
