Summary: In this study, a multicenter retrospective analysis was conducted to investigate cases with specific criteria related to germ cell tumors (GCTs). The results showed that all patients meeting the criteria had GCTs or granulomatous inflammation, not other types of tumors. Biopsy was recommended, considering the presence of non-germinomatous GCTs in at least 3.4% of the patients and the safety and efficacy of simultaneous procedures for acute hydrocephalus.

Summary: The study shows that GLI3 is highly expressed in both WNT-activated and SHH-activated medulloblastoma, with GLI1 and GLI3 being highly expressed in SHH-activated medulloblastoma. In contrast, GLI3 is significantly expressed in WNT-activated medulloblastoma while GLI1 is suppressed.

Summary: Recent studies have shown the feasibility of detecting H3K27M mutations in diffuse midline glioma patients' cerebrospinal fluid. However, liquid biopsy only achieved a definitive detection of the mutation in a small percentage of patients. Younger age and higher concentration of CSF protein tended to be associated with cases where a definitive assessment was possible.

Summary: Detecting BRAF V600E mutation in GBMs is crucial due to potential treatment implications, however, the scarcity of these mutations poses challenges for molecular testing in all GBMs. Analyzing clinical, radiographic, and pathologic features of BRAF V600E-mutant GBM compared to controls revealed specific MRI characteristics, higher likelihood of total resection, and distinctive epithelioid features, emphasizing the importance of testing for BRAF V600E in IDH-wildtype GBM.

Survival prediction based on the gene expression associated with cancer morphology and microenvironment in primary central nervous system lymphoma

Summary: Dysregulation of cell morphology and cell-cell interaction is crucial for cancer cell growth and metastasis. The balance between ECM and MMP plays a key role in cancer cell morphology and angiogenesis. Gene signatures associated with PCNSL morphology and microenvironment were identified through NGS, with prognostic prediction formulas developed and collagen genes found to contribute to microenvironment regulation. A combinatorial expression of representative genes enabled precise prognosis prediction and diagnosis of PCNSL cell types with MTX resistance.

Summary: The study investigated expression modules reflecting the reciprocal expression of cancer microenvironment and immune response-related genes associated with poor prognosis in primary central nervous system lymphoma (PCNSL). Weighted gene coexpression network analysis revealed representative modules including neurogenesis, immune response, anti-virus, microenvironment, gene expression, and translation, extracellular matrix, morphogenesis, and cell adhesion. The results suggest that the differential expression and balance of immune response and microenvironment genes may play a crucial role in the growth and prognosis prediction of PCNSL tumors.
