Mode of Anisotropy Reveals Global Diffusion Alterations in Attention-Deficit/Hyperactivity Disorder

Objective: Diffusion tensor imaging (DTI) can identify structural connectivity alterations in attention-deficit/hyperactivity disorder (ADHD). Most ADHD DTI studies have concentrated on regional differences in fractional anisotropy (FA) despite its limited sensitivity to complex white matter architecture and increasing evidence of global brain differences in ADHD. Here, we examine multiple DTI metrics in separate samples of children and adults with and without ADHD with a principal focus on global between-group differences. Method: Two samples: adults with ADHD (n = 42) and without (n = 65) and children with ADHD (n = 82) and without (n = 80) were separately group matched for age, sex, and head motion. Five DTI metrics (FA, axial diffusivity, radial diffusivity, mean diffusivity, and mode of anisotropy) were analyzed via tract-based spatial statistics. Group analyses tested for diagnostic differences at the global (averaged across the entire white matter skeleton) and regional level for each metric. Results: Robust global group differences in diffusion indices were found in adults, with the largest effect size for mode of anisotropy (MA; Cohen's d = 1.45). Global MA also differed significantly between groups in the pediatric sample (d = 0.68). In both samples, global MA increased classification accuracy compared to the model with clinical Conners' ADHD ratings alone. Regional diagnostic differences did not survive family wise correction for multiple comparisons. Conclusion: Global DTI metrics, particularly the mode of anisotropy, which is sensitive to crossing fibers, capture connectivity abnormalities in ADHD across both pediatric and adult samples. These findings highlight potential diffuse white matter microarchitecture differences in ADHD.

Summary: Males exhibit greater variability than females in many traits, which may have implications for understanding sex differences in health and disease. A mega-analysis of brain structure variability based on MRI data of 16,683 healthy individuals spanning nine decades of life reveals significant patterns of greater male than female between-subject variance in subcortical volumes, cortical surface area, and cortical thickness. These sex differences are present in childhood and may be influenced by early life genetic or gene-environment interaction mechanisms. The findings underscore the importance of individual differences within each sex, which may contribute to sex-specific vulnerability to disorders.

Reproducibility in the absence of selective reporting: An illustration from large-scale brain asymmetry research

Summary: The problem of poor reproducibility of scientific findings has been a topic of concern in recent years. A recent multisite collaborative study investigated the reproducibility of brain anatomical left-right asymmetries in regional measures of surface area and cortical thickness. The study found an average reproducibility rate of 63.2%, with larger effects and larger datasets having higher reproducibility.

Summary: The ability of automatic feature learning in Convolutional Neural Network (CNN) is potentially suitable for uncovering complex and widespread brain changes in schizophrenia. This study developed a deep feature approach using 2D and 3D CNN models to classify schizophrenia based on multimodal neuroimaging data. The 3D CNN models outperformed the 2D CNN models and handcrafted feature-based machine learning approach, and identified critical brain regions for illness classification.

Multimodal gradient mapping of rodent hippocampus

Summary: The hippocampus plays a crucial role in our sense of self and our place in the world. Through gradient analysis on resting state fMRI data, we found two major functional connectivity gradients in the hippocampus. One showed a sharp discontinuity, while the other was more continuous. The dorsal region was more strongly connected to areas involved in spatial navigation, while the ventral region was more strongly connected to areas involved in emotion. Gene co-expression gradients, however, showed a more segregated and discrete organization.

Self-Supervised Graphs for Audio Representation Learning With Limited Labeled Data

Summary: This paper introduces a self-supervised graph approach for learning audio representations from limited labelled data. By constructing subgraphs and utilizing self-supervision tasks, effective audio representations can be learned. Decreased basal ganglia and thalamic iron in early psychotic spectrum disorders are associated with increased psychotic and schizotypal symptoms

Summary: This study used quantitative MRI to compare iron content in several iron-rich subcortical structures between individuals with PSD and healthy controls. The results showed significantly reduced iron content in PSD patients compared to controls, particularly in patients with schizophrenia. These findings suggest that decreased iron in subcortical regions is associated with PSD risk and symptomatology, and may play a role in the pathology of PSD.

Rethinking academia in a time of climate crisis

Summary: Addressing the climate crisis requires radical and urgent action at all levels of society. Universities, in particular, have a responsibility to take the lead in such action, but they are falling short. Academic scientists also face obstacles such as bureaucracy and excessive competition, hindering their work. Drawing on Doughnut Economics, we propose new principles for scientific practice and urge academics to create a scientific enterprise that is capable of meeting the challenges of the 21st century.

White matter alterations in Attention-Deficit/Hyperactivity Disorder (ADHD): a systematic review of 129 diffusion imaging studies with meta-analysis

Summary: This study conducted a systematic review and meta-analysis of diffusion weighted imaging (DWI) studies to investigate abnormal anatomical brain connections in ADHD. The findings show white matter alterations in projection, commissural and association pathways, especially reduced fractional anisotropy (FA), which are associated with symptom severity and cognitive deficits. Despite some methodological limitations, these findings have important implications for understanding the brain effects of ADHD and guiding clinical interventions.

Mindfulness-based therapy improves brain functional network reconfiguration efficiency

Summary: Mindfulness-based interventions have shown promise in treating psychological disorders by improving cognition and emotion regulation. Understanding the underlying neural plasticity and functional brain changes associated with mindfulness interventions is important for current neuroimaging research. Previous studies have found functional brain changes during resting and task states, particularly in executive control, default mode, and salience networks. However, limited research has combined information from rest and task to study mindfulness-related functional changes, especially in the context of intervention studies with active controls. Recent work has discovered that the reconfiguration efficiency of brain activity patterns between rest and task is behaviorally relevant. In this study, we applied this measure to investigate how mindfulness intervention influenced functional reconfiguration between rest and a breath-counting task in elderly participants with sleep difficulties. By comparing the effects of a mindfulness-based therapy and an active control intervention, we found that mindfulness intervention improved self-reported mindfulness measures and brain functional reconfiguration efficiency in executive control, default mode, and salience networks. However, the brain and behavioral changes were not associated with each other. These findings suggest that regular mindfulness practice may induce neuroplasticity, bringing the intrinsic functional configuration of participants' brains closer to a state required for mindful awareness.
