Summary: In this study, an automated liquid handling workstation was developed for the density-based separation of extracellular vesicles (EV) from human body fluids. The performance of the automated method was compared to manual handling by experienced or inexperienced researchers. The results showed that the automated EV separation method significantly reduced the variability in EV recovery and exhibited high reproducibility, specificity, and reduced hands-on time.

Summary: A novel 3D printing leukocyte sorting accumulator combined with centrifugation was demonstrated to achieve label-free initial leukocyte enrichment based on cell density and size. The upgraded design, leukocyte sorting accumulator-3, optimized the recovery of buffy coats compared to earlier models, with mean leukocyte recovery rates of 96.2 +/- 2.38% by pushing method and 94.4 +/- 0.8% by pulling method. New procedures for preliminary leukocyte enrichment are essential for modern clinical and basic research assays.

Summary: A method for monitoring the efficiency of separating hybrid magnetoliposomes using multiphase density gradient centrifugation coupled with a continuous flow system is described. Different h-MLs suspensions containing hydrophobic magnetic gold nanoparticles and various fluorophores were synthesized and separated using the MDGC system. The separation efficiency was tested and different fractions were identified for each separated h-MLs sample.

Summary: The separation of blood plasma by centrifugation can be accelerated by tilting the sample relative to the direction of the centrifugal force. This is due to the Boycott effect and buoyancy-induced convection, resulting in enhanced sedimentation of red blood cells. Flow instability invalidates the traditional predictive model, but our new model accounts for these factors and predicts the optimal tilt angle for highest separation efficiency. Experimental data confirms the accuracy of our theoretical prediction, and sensitivity analysis investigates the influence of blood sample variation on physical parameters.

Summary: This study compares the effects of classic density-gradient purification and immunomagnetic isolation on the phenotype and functionality of neutrophils. The results show that the two methods produce cells with different characteristics under different experimental conditions. Density-gradient-derived neutrophils exhibit increased polarization responses and release of reactive oxygen species, neutrophil extracellular traps, and granular proteins, while immunomagnetic isolation-derived neutrophils are mostly quiescent. The choice of purification method should depend on the specific experimental purposes.

Summary: This study compared three commonly used methods, swim-up, Percoll gradient, and washing by centrifugation, for extracting sperm from collared peccaries. The results showed that swim-up method had poor efficiency in isolating motile sperm, while Percoll gradient and washing by centrifugation had similar quality and fertilization capacity of the sperm. Therefore, Percoll gradient and washing by centrifugation are recommended for sperm extraction in collared peccaries.

Summary: Flow cytometry has been used to understand the impact of viruses on shaping marine microbial communities and biogeochemical cycles in the pelagic zone. A new protocol has been developed for the enumeration of marine sediment viruses by flow cytometry, enabling fast and accurate quantification of viral abundances and opening up options for virus sorting, targeted viromics, and single-virus sequencing. Further optimization may be needed for sediments with low biomass or high organic content.
