Missense VKOR mutants exhibit severe warfarin resistance but lack VKCFD via shifting to an aberrantly reduced state

The study demonstrates that missense mutations in the VKOR gene cause resistance to warfarin treatment without abnormal bleeding. The redox state of the mutants plays a critical role in their activity and warfarin resistance.
Missense vitamin K epoxide reductase (VKOR) mutations in patients cause resistance to warfarin treatment but not abnormal bleeding due to defective VKOR activity. The underlying mechanism of these phenotypes remains unknown. Here we show that the redox state of these mutants is essential to their activity and warfarin resistance. Using a mass spectrometry-based footprinting method, we found that severe warfarin-resistant mutations change the VKOR active site to an aberrantly reduced state in cells. Molecular dynamics simulation based on our recent crystal structures of VKOR reveals that these mutations induce an artificial opening of the protein conformation that increases access of small molecules, enabling them to reduce the active site and generating constitutive activity uninhibited by warfarin. Increased activity also compensates for the weakened substrate binding caused by these mutations, thereby maintaining normal VKOR function. The uninhibited nature of severe resistance mutations suggests that patients showing signs of such mutations should be treated by alternative anticoagulation strategies.

Study on the minimum extinguishing concentration of C6F12O for extinguishing synthesis gas flame of lithium-ion battery

Summary: In this study, a liquid-phase input end is added to the cup burner to test the fire-extinguishing effect of C6F12O on lithium-ion battery syngas. The minimum extinguishing concentration (MEC) of C6F12O is determined, and the physical and chemical effects of C6F12O on lithium-ion battery syngas fire are quantified using the perfect stirred reactor (PSR) model. The results indicate that the fire-extinguishing effect of C6F12O on lithium-ion battery fire is mainly due to its physical cooling effect.

Prediction of Photochemical Properties of Dissolved Organic Matter Using Machine Learning

Summary: Machine learning models were developed to predict the values of three photochemical reactive intermediates (PPRIs) apparent quantum yields (phi) based on dissolved organic matter (DOM) spectral parameters, experimental conditions, and calculation parameters. The CatBoost model showed the best predictive performance, outperforming traditional linear regression models. The significance of wavelength range and spectral parameters on phi PPRIs predictions was revealed, indicating that DOM with lower molecular weight, lower aromatic content, and a more autochthonous portion possessed higher phi PPRIs.

Description of age-related changes in cervical sagittal alignment based on pelvic incidence classification in asymptomatic Chinese population

Summary: The purpose of this study was to investigate the PI- and age-related cervical alignment changes of Chinese healthy population. The results showed that there was no obvious difference observed between PI and cervical sagittal parameters, except for caudal arch. However, C2-7, cranial arch, caudal arch, T1S, and C2-7 SVA increased remarkably with age. Based on the classification in the study, high or low PI apparently did not correlate with the occurrence of cervical degenerative disease.

Summary: The long noncoding RNA (lncRNA) MetBil plays a significant role in cardiac fibrosis by interacting with METTL3 and regulating the expression of methylated fibrosis-associated genes. Overexpression of MetBil enhances collagen deposition, cardiac fibroblast proliferation, and activation, while silencing MetBil has the opposite effects. This study provides a new potential target for interventions in fibrosis-related cardiac diseases.

Influence of flue gas components on the mercury adsorption/oxidation by mechanochemical S2Cl2-modified sawdust coke

Summary: This study investigated the effects of flue gas components (O2, CO2, SO2, and NO) on the Hg-0 removal of mechanochemical S2Cl2-modified BC (S2Cl2-MC). It was found that O2 and CO2 promoted the escape of Hg2+ but inhibited the adsorption of Hg-0, while SO2 and NO had a certain promoting effect on the removal of Hg-0.

Gene expression of TRPMLs and its regulation by pathogen stimulation

Summary: The three members of TRPML subfamily, TRPML1, TRPML2, and TRPML3, are highly expressed in the lung tissue, as well as spleen and kidney tissues of mice. TRPML1 and TRPML3 showed decreased expression in response to LPS stimulation, while TRPML2 showed increased expression. This suggests that TRPML1 and TRPML3 play important roles in immune and inflammatory regulation.

A Bibliometric and Visualized Analysis of Remote Sensing Methods for Glacier Mass Balance Research

The research findings provide valuable insights for future studies and highlight the importance of improving data quality and merging multiple methods for precise and long-term monitoring of glacier mass changes.

Au/CeO2 NR Restricted Inside Cu-MOFs: A Three-in-One Artificial Enzyme with Synergistically Enhanced Peroxidase-Like Activity for Dual-Mode Sensing of Multiple Biomarkers

Summary: In this work, a three-in-one artificial enzyme with enhanced peroxidase-like activity and outstanding stability is constructed by restricting Au-loaded CeO2 nanorods inside Cu-based metal-organic frameworks. The artificial enzyme shows significantly improved catalytic decomposition of H2O2 and strong affinities to substrates, with lower Michaelis constants than horseradish peroxidase. It also exhibits desirable stabilities in pH, temperature, organic solvents, and long-term storage. A universal colorimetric/fluorometric dual-mode sensing platform is built based on the artificial enzyme for detection of biomarkers in biological fluids. This work opens up new possibilities for designing efficient and stable biomimetic catalysts.

Pd-Catalyzed Dehydrative Synthesis of Allylic Amines in Water

A novel gene REPTOR2 activates the autophagic degradation of wing disc in pea aphid

Summary: Wing dimorphism in insects is an evolutionarily adaptive trait, with wingless individuals undergoing wing degeneration via autophagy. The TOR signaling pathway suppresses autophagy of the wing disc by negatively regulating the expression of the gene REPTOR2. Knockdown of REPTOR2 reduces autophagy and increases the proportion of winged individuals, while suppression of TOR activates autophagy and decreases the proportion of winged individuals. REPTOR2 acts as a key regulator downstream of TOR in the signaling pathway.

Altered gut microbiome profile in patients with knee osteoarthritis

Summary: In this study, the gut microbiota profile in patients with knee osteoarthritis (OA) was examined using 16S rDNA gene sequencing and metagenomic sequencing. The dominant bacteria in the OA group was identified as Proteobacteria and 81 genera showed significantly different abundance between the OA and control groups. The metagenomic sequencing revealed specific species, such as Bacteroides stercoris and Escherichia coli, that were significantly altered in abundance in the OA group. These findings suggest a potential role of the cartilage-gut-microbiome axis in the mechanisms and pathogenesis of OA.
