Radiogenomics of diffuse intrinsic pontine gliomas (DIPGs): correlation of histological and biological characteristics with multimodal MRI features

Summary: This study analyzed the MRI and histopathological features of 27 children with DIPG H3K27M mutant tumors, revealing that H3.1K27M mutated tumors have higher ADC values and lower perfusion values compared to H3.3K27M ones, possibly due to gated channel activity-related gene expression.

A DNA Repair and Cell Cycle Gene Expression Signature in Pediatric High-Grade Gliomas: Prognostic and Therapeutic Value

Natacha Entz-Werle, Laetitia Poidevin, Petr V. Nazarov, Olivier Poch, Benoit Lhermitte, Marie Pierre Chenard, Helene Burckel, Eric Guerin, Quentin Fuchs, David Castel, Georges Noel, Laurence Choulier, Monique Dontenwill, Eric Van Dyck

Summary: Pediatric high-grade gliomas (pHGGs) are incurable brain tumors that exhibit resistance to standard therapies. This study investigated the expression of DNA repair and cell cycle genes in pHGGs to propose a novel classification based on therapeutic vulnerabilities. Several potential new diagnostic markers were identified among the DNA repair factors studied.

Summary: This study identified three unclassified vascular lesions in the central nervous system (CNS) that shared radiological and histopathological features similar to a subtype of angioleiomyomas in soft tissue. Molecular analysis revealed a common GJA4 mutation in these lesions, which was also found in soft tissue angioleiomyomas. Further analysis showed that these CNS lesions formed a distinct epigenetic group separate from other vascular tumors and soft tissue angioleiomyomas.

The genomic landscape of dysembryoplastic neuroepithelial tumours and a comprehensive analysis of recurrent cases

Jones, Raphael Saffroy, David Castel, Thomas Blauwblomme, Alice Metais, Marie Bourgeois, Emmanuele Lechapt-Zalcman, Arnault TauziedeEspariat, Felipe Andreiuolo, Fabrice Chretien, Jacques Grill, Nathalie Boddaert, Dominique Figarella-Branger, Rameen Beroukhim, Pascale Varlet

Summary: Dysembryoplastic neuroepithelial tumour (DNT) is a challenging glioneuronal tumour with diverse histological features. The study used comprehensive analysis techniques to characterize 82 DNTs, revealing differences between specific and non-specific/diffuse DNTs, as well as identifying additional genomic alterations in a subset of DNTs.

Posterior fossa ependymoma H3 K27-mutant: an integrated radiological and histomolecular tumor analysis

Summary: This study retrospectively investigated 9 cases of pediatric H3 K27M-mutant EPN_PFA and found that 7 tumors clustered with EPN_PFA, while one was reclassified as DMG. H3 K27M-mutant EPN_PFA cases were older and showed similarities in radiology and histopathology with EZHIP-overexpressing EPN_PFA. Sequencing analysis revealed K27M mutations in HIST1H3B, HIST1H3C, H3F3A, and HIST1H3D. Overall survival rates were similar between EZHIP-overexpressing and H3 K27M-mutant EPN_PFA.

Summary: A study identified a specific molecular class of pediatric-type malignant gliomas with EGFR amplifications or mutations, as well as H3.1 or H3.3 K27M mutations, all showing loss of H3K27me3 staining. While some tumors exhibited a bithalamic growth pattern, a significant proportion occurred in other locations.
