Summary: This study systematically analyzed the functions of long non-coding RNAs (lncRNAs) at the single-cell level in tumor immune microenvironments (TIMEs). A tool called scLNC was developed for the functional characterization of lncRNAs at the single-cell level. The study identified cell-type-specific lncRNA functions and revealed new regulatory units established by lncRNAs in tumor-infiltrating T cells, particularly in tumor-enriched T cells. Overall, lncRNAs were found to play an important role in regulating cytokine production, cell activation, and migration in tumor-enriched T cells, which has implications for immunotherapy.
