Acid-sensing ion channels (ASICs) 2 and 4.2 are expressed in the retina of the adult zebrafish

Acid-sensing ion channels (ASICs) are H+-gated, voltage-insensitive cation channels involved in synaptic transmission, mechanosensation and nociception. Different ASICs have been detected in the retina of mammals but it is not known whether they are expressed in adult zebrafish, a commonly used animal model to study the retina in both normal and pathological conditions. We study the expression and distribution of ASIC2 and ASIC4 in the retina of adult zebrafish and its regulation by light using PCR, in situ hybridization, western blot and immunohistochemistry. We detected mRNA encoding zASIC2 and zASIC4.2 but not zASIC4.1. ASIC2, at the mRNA or protein level, was detected in the outer nuclear layer, the outer plexiform layer, the inner plexiform layer, the retinal ganglion cell layer and the optic nerve. ASIC4 was expressed in the photoreceptors layer and to a lesser extent in the retinal ganglion cell layer. Furthermore, the expression of both ASIC2 and ASIC4.2 was down-regulated by light and darkness. These results are the first demonstration that ASIC2 and ASIC4 are expressed in the adult zebrafish retina and suggest that zebrafish could be used as a model organism for studying retinal pathologies involving ASICs.

A Promising Antifungal and Antiamoebic Effect of Silver Nanorings, a Novel Type of AgNP

Summary: Silver nanoparticles have potent antimicrobial activity and are investigated as an alternative to antimicrobial drugs. This study analyzed the antifungal and antiamoebic effects of a new type of silver nanoparticles called silver nanorings (AgNRs), and compared them with other types of silver nanoparticles. The results showed that silver nanoparticles have high biocidal effect, with antifungal activity depending on the fungal species involved, and AgNRs and silver nanowires (AgNWs) having an inhibitory effect on the growth of Acanthamoeba castellanii.

Summary: This study characterizes the leukocytes present in the gut-associated lymphoid tissue (GALT) of H. niloticus for the first time using confocal immuno-fluorescence techniques. This expands our understanding of intestinal immunity in this fish.

Zebrafish: A Model Deciphering the Impact of Flavonoids on Neurodegenerative Disorders

Summary: In the past century, advancements in biotechnology, biochemistry, and pharmacognosy have brought attention to flavonoids, which are polyphenolic secondary metabolites capable of modulating various biological mechanisms, including those involved in neuronal plasticity, learning, and memory. Flavonoids also impact biological processes associated with neurodegenerative diseases such as oxidative stress, neuroinflammation, and mitochondrial dysfunction. Therefore, certain flavonoids can be used as adjuvants to prevent and counteract disorders like Alzheimer's and Parkinson's diseases. Zebrafish, with its high genome homology to humans, similar brain organization and neurochemical processes, and maintained neurogenic activity in the adult brain, provides a valuable model for studying human neurodegenerative diseases and the effects of flavonoids on these disorders.

PINK1 Immunoexpression Predicts Survival in Patients Undergoing Hepatic Resection for Colorectal Liver Metastases

Summary: The study investigated the expression of PTEN-induced kinase-1 (PINK1) in surgical specimens from ninety patients with metastatic colorectal adenocarcinoma (CRC) to the liver. The results showed that positive PINK1 expression was observed in all tumoral tissues, and there were no differences between different CRCs. However, a significant under-expression of PINK1 was found in CRLMs derived from the rectum compared to other sites. Altered PINK1 expression in CRCs was associated with worse overall survival due to abnormal mitophagy function.

Summary: The recognition and elimination of invading pathogens are essential for host survival. Macrophages, which function similarly to vertebrate macrophages, play a crucial role in host protection in all multicellular organisms. CD14 is a pattern responsible for bacterial recognition found on the surface of macrophages. These cells store antimicrobial molecules and display polarization states similar to mammalian macrophages. The present study reports the presence of various cell markers, including CD14, in the intestinal macrophages of catfish and bonytongue, providing insights into the in vivo function of macrophages in fish.
