Evidence of DMSO-Induced Protein Aggregation in Cells

We report on a study of protein aggregation induced on different cell samples by dimethyl sulfoxide (DMSO) addition. DMSO is the most commonly used cryoprotectant because it is supposed to readily diffuse across lipid bilayers, thus reducing water activity within cells; despite its large use, the mechanism of penetration and even the main interaction features with cell components are far from being understood. In the present work, infrared absorption spectroscopy is successfully applied to real time detection of chemical and structural changes occurring in cells during dehydration from water and water/DMSO suspensions. As a most interesting result, DMSO is observed to favor protein aggregation both in cellular model systems, as cultured lymphocytes and fibroblasts, and in human samples for clinic use, as hematopoietic stem cells from cord blood. This effect is evidenced at low water content, analogously to what is observed for protein solutions. Such tendency is not specific of the type of protein and suggests one possible origin of DMSO toxicity.

Summary: This study aims to investigate the lipidomic analysis of erythrocytes in order to highlight variations between healthy and patient subjects. The results showed that the lipid composition of sickle cells, especially phospholipids, were altered in SCD patients. In-depth study of changes in lipid metabolism can provide insights into the enzymes and genes involved, improving the understanding of the biochemical basis of SCD and potentially developing a predictive score system for clinical severity.

Extracellular Vesicles in Aging: An Emerging Hallmark?

Summary: Extracellular vesicles (EVs) are membrane-enclosed particles secreted by cells and circulating in body fluids. They not only serve as a tool for waste disposal, but also transmit cell signals. Aging is characterized by genomic instability, telomere erosion, epigenetic alteration, and defective proteostasis. Studies have shown that EVs released in cell models are associated with these aging hallmarks and can spread senescence signals. Furthermore, EVs have great potential as anti-aging tools. Although this is a recent field, altered EV secretion in aging suggests it as an emerging hallmark of aging.

Recent advances in lipid analysis have allowed for the investigation of the lipid content in cancer patient biofluids, leading to the discovery of potential cancer biomarkers. Biofluid-derived extracellular vesicles, with their unique molecular signature, have emerged as a promising platform for lipid biomarker discovery.

Summary: The characterization of the protein and lipid content of milk extracellular vesicles (EVs) from different mammal species is crucial for understanding their biogenesis and biological functions, as well as for a comprehensive description of the nutritional aspects of animal milk for human diet. Limited studies have been conducted on the protein and lipid composition of milk EVs compared to those investigating nucleic acid cargo. However, recent research suggests that the biochemical cargo of milk EVs differs from other milk fractions, and there are variations in the composition of milk EVs among different animal species and under different lactation stages and health statuses.

Use of Biomass Ash in Reinforced Clayey Soil: A Multiscale Analysis of Solid-State Reactions

Summary: The use of biomass ashes as stabilizing agents can improve the mechanical properties of clayey soils. The binding agent production process is energy-intensive and emits large amounts of CO2. Through the analysis of the chemical and mineralogical changes in clay-ash mixtures and mechanical tests, it was found that the dissolution of clay minerals and the reaction of the amorphous phase in the ash contribute to the stiffening and strengthening of clayey soils.

FTIR Analysis of Renal Tissue for the Assessment of Hypertensive Organ Damage and proANP31-67 Treatment

Summary: The kidneys play an important role in the regulation of high blood pressure, but the mechanisms behind renal damage in hypertension are still being investigated. Fourier-Transform Infrared (FTIR) micro-imaging was used to monitor the early biochemical changes in salt-induced hypertension in rats. FTIR imaging and principal component analysis detected hypertension-induced alterations in the renal parenchyma and blood vessels. Amino acids and protein contents in renal blood vessels were independent of lipid, carbohydrate, and glycoprotein contents in the renal parenchyma. FTIR micro-imaging is a reliable tool for monitoring kidney tissue and its hypertension-induced alterations, and it detected a significant reduction in these alterations in proANP(31-67)-treated rats, suggesting the beneficial effects of this novel medication on the kidneys.

Summary: Telomeric G-quadruplexes (G4s) exhibit structural polymorphism and the dynamics of G4s are affected by conformation and complexation. In this study, the dynamics of Tel22 in different conformations and complexed with BRACO19 ligand were investigated. The results showed that hydration water played a role in mediating the effect of polymorphism and complexation on the fast dynamics of G4s.

Effect of plasmonic excitation on mature insulin amyloid fibrils

Summary: In this study, gold nanoparticles were used as photothermal agents to alter the morphology of insulin fibrils. The effects on the structure of mature insulin fibrils upon plasmonic excitation of nanoparticles were monitored. The results indicate that amyloid aggregates can be effectively destroyed by irradiation of plasmonic nanoparticles, suggesting potential strategies for modifying the structure of amyloid fibrils.

Extracorporeal membrane oxygenation and effects on tendon tissue: A vibrational spectroscopy study

Summary: In this study, Raman microspectroscopy and attenuated total reflection-Fourier transform infrared micro-imaging were used to analyze samples of tibial and peroneal human tendons from ECMO and non-ECMO donors. The results showed significant differences in the chemical composition and extracellular matrix characteristics between tendons from these two groups, indicating changes in amino acid content and protein structure.

Electrospun Nanofibrous UV Filters with Bidirectional Actuation Properties Based on Salmon Sperm DNA/Silk Fibroin for Biomedical Applications

Summary: In this study, a bioinspired spinning process was used to produce an electrospun nanofibrous silk-based patch. The patch demonstrated bidirectional macroscale actuation behavior in response to water vapor and displayed excellent UV-blocking properties. By incorporating DNA, the patch showed enhanced electrical conductivity and water-insolubility. Biological experiments confirmed that the patch provided photoprotection and improved cellular viability after UV exposure. Moreover, the patch treated with water vapor exhibited potential application as a scaffold for tissue engineering and promoted tissue regeneration when combined with HaCaT cells.

Summary: HFpEF is a complex cardiovascular disease with unknown progression mechanism and challenging diagnosis. This study used FTIR and Raman spectroscopies to detect biochemical alterations related to HFpEF and its associated comorbidities in the rat ventricles, demonstrating the high sensitivity and selectivity of these techniques in detecting changes in heart chemistry.
