Summary: The loss of TP53 and RB1 in treatment-naive small cell lung cancer (SCLC) indicates a need to inactivate cell death pathways prior to therapy. This study identifies non-neuroendocrine (NE) SCLC as vulnerable to ferroptosis and neuroendocrine (NE) SCLC as susceptible to TRX pathway inhibition, with combining these treatments showing promise in overcoming subtype plasticity in SCLC.

Summary: This study investigated the effects of c-FLIP on the expression and ubiquitination levels of FoxM1 in non-small-cell lung cancer (NSCLC) cells, and explored the role of c-FLIP/FoxM1 in drug resistance. The findings demonstrated that c-FLIP stabilized FoxM1 by inhibiting its ubiquitination, and promoted the resistance of NSCLC cells to thiostrepton and osimertinib by upregulating FoxM1. The combination of FoxM1 and c-FLIP was found to be a more accurate prognostic biomarker for NSCLC patients.

The results showed that the prognosis and treatment modalities were similar between these two types of advanced lung cancers. Chemoradiotherapy significantly improved survival in both LCNEC and SCLC patients.

Summary: The research found that miR-22-3p, miR-9-5p, and miR-218-5p are antiproliferative miRNAs targeting non-small cell lung cancer (NSCLC) cells, and they inhibit folate metabolism and alter amino acid metabolism in NSCLC cells. In addition, the inhibition of glucose uptake by miR-22-3p and the reduction of serine biosynthesis from glucose by miR-9-5p and -218-5p were also confirmed. Recombinant miR-22-3p was more effective than miR-9-5p and -218-5p in inhibiting NSCLC cell respiration, glycolysis, and colony formation, without causing any toxicity.

Summary: Mucin4 (MUC4) expression coincides with EGFR expression in early PanIN1 lesions, and sustained EGFR signaling in PanIN lesions is dependent on MUC4. Depletion of MUC4 delays PanIN lesion formation and reduces EGFR phosphorylation, suggesting impaired oncogenic signaling. MUC4 interacts with EGFR and prevents its degradation, leading to sustained downstream signaling. Targeting the MUC4-EGFR interaction may enhance the efficacy of EGFR-targeted therapies in PDAC.
