Radio-protective effect of some new curcumin analogues

In the present study, novel symmetrical curcumin analogues (2-7) have been synthesized by substituting the phenolic OH of curcumin with different linkers providing additional keto-enol tautomerism, very essential for radioprotective activity. The structures of the synthesized compounds (2-7) were elucidated by elemental analysis, IR, H-1-NMR, C-13-NMR and mass spectral data and were found consistent with the assigned structures. The curative effect of these new compounds, against the oxidative stress due to exposure of rats to the whole body gamma-irradiation (7 Gy) was investigated. Gamma-irradiated rats exhibited elevations of ALT, AST activities, urea, creatinine, triglycerides, total cholesterol, malondialdehyde (MDA), nitric oxide (NO), Interleukin-6 (IL-6), Tumor Necrosis Factor-alpha (TNF-alpha) and Nuclear Factor-kappa B (NF-kappa B) levels. Contrariwise, the total protein, albumin, total calcium level, SOD, CAT, GSH-Px, GST activities and GSH content were decreased. Treatment of gamma-irradiated rats with the new curcumin analogues (2-7) showed significant amelioration in the in-vivo antioxidant status, liver and kidney functions, as well as the anti-inflammatory markers (IL-6, TNF-alpha and NF-kappa B). Inhibition of NF-kappa B could be responsible for the improvement of the antioxidant and anti-inflammatory status in gamma-irradiated animals, by down-regulation of IL-1 beta and TNF-alpha level. In conclusion, the new curcumin analogues (2-7) exhibited post-protective effect on gamma-irradiation, by NF-kappa B inhibition. (C) 2016 Elsevier B.V. All rights reserved.

Quinazolinone derivatives as new potential CDK4/6 inhibitors, apoptosis inducers and radiosensitizers for breast cancer

Summary: New quinazolinones with acetamide linkers were synthesized as potential anti-breast cancer agents. In vitro cytotoxic evaluation identified 1,3-benzodioxole (5d) as the most potent compound with good inhibitory activity on CDK4/6. Compound 5d arrested the cell cycle, induced apoptosis, and showed good radiosensitizing activity when combined with γ-radiation in breast cancer cells. This study introduces quinazolinone scaffolds as new CDK4/6 inhibitors for breast cancer treatment.

Rutin protects against gamma-irradiation and malathion-induced oxidative stress and inflammation through regulation of mir-129-3p, mir-200C-3p, and mir-210 gene expressions in rats' kidney

Summary: This study investigated the protective response of rutin against kidney injury induced by malathion and gamma-irradiation in rats. The results showed that rutin treatment regulated various biochemical parameters, oxidative stress, antioxidant status, and inflammatory responses in the kidney tissues. It also modulated the activity of enzymes, gene expressions, and protein expressions related to renal function. Histopathological examinations confirmed these findings.

Amplifying bactericidal activity: Surfactant-mediated AgBr thin film coating over two-dimensional vertically aligned ZnO nanorods for dark-light dual mode disinfection

Summary: Thin film coatings with potent antibacterial properties are widely used in medical devices, frequently touched surfaces, and food packaging. However, achieving antibacterial performance under both light and dark conditions remains a challenge. In this study, we successfully prepared AgBr-coated ZnO nanorod thin film architecture using a unique surfactant-mediated solution-phase spin-coating approach. The resulting structures showed dual functionality as antimicrobial agents and photosensitizers, effectively enhancing the visible-light photodisinfection efficacy of ZnO nanorods. The concentration of the surfactant-based precursor solution could be modulated to control the AgBr content. Additionally, components like urea and lactic acid were found to enhance the photocatalytic efficiency of the structures.
