Fatty Acid-Binding Protein 5 at the Blood-Brain Barrier Regulates Endogenous Brain Docosahexaenoic Acid Levels and Cognitive Function

Fatty acid-binding protein 5 (FABP5) at the blood-brain barrier contributes to the brain uptake of docosahexaenoic acid (DHA), a blood-derived polyunsaturated fatty acid essential for maintenance of cognitive function. Given the importance of DHA in cognition, the aim of this study was to investigate whether deletion of FABP5 results in cognitive dysfunction and whether this is associated with reduced brain endothelial cell uptake of exogenous DHA and subsequent attenuation in the brain levels of endogenous DHA. Cognitive function was assessed in male and female FABP5(+/+) and FABP5(-/-) mice using a battery of memory paradigms. FABP5(-/-) mice exhibited impaired working memory and short-term memory, and these cognitive deficits were associated with a 14.7 +/- 5.7% reduction in endogenous brain DHA levels. The role of FABP5 in the blood-brain barrier transport of DHA was assessed by measuring C-14-DHA uptake into brain endothelial cells and capillaries isolated from FABP5(+/+) and FABP5(-/-) mice. In line with a crucial role of FABP5 in the brain uptake of DHA, C-14-DHA uptake into brain endothelial cells and brain capillaries of FABP5(-/-) mice was reduced by 48.4 +/- 14.5% and 14.0 +/- 4.2%, respectively, relative to those of FABP5(+/+) mice. These results strongly support the hypothesis that FABP5 is essential for maintaining brain endothelial cell uptake of DHA, and that cognitive deficits observed in FABP5(-/-) mice are associated with reduced CNS access of DHA.

Lipase-catalysed changes in essential oils revealed by comprehensive two-dimensional gas chromatography

Summary: Candida antarctica lipase A (CALA) was used for chemo-selective enzymatic transesterification of terpene and phenyl alcohols in 35 different essential oil samples. The results showed high conversion rates for primary and secondary alcohols, while tertiary alcohols and phenols showed no significant conversion. The process demonstrated selectivity, efficiency, scalability, and potential reusability, making it a greener and more sustainable alternative for the production of novel flavors and fragrances in the industry.

Development and Application of a Novel Multiloop Splitter-Based Non-cryogenic Artificial Trapping Modulation System in Comprehensive Two-Dimensional Gas Chromatography

Summary: A non-cryogenic artificial trapping modulation technique, called M-SNAT, was developed using multiloop splitters and various columns. This technique achieved artificial splitting and trapping of analytes by utilizing split pulses, pulse delay, and their combination within each loop. The SNAT approach was successfully applied for modulating a wide range of compounds in waste tire pyrolysis samples, resulting in high precision and improved separation capacity.

Peripheral Administration of the Kv1.3-Blocking Peptide HsTX1[R14A] Improves Cognitive Performance in Senescence Accelerated SAMP8 Mice

Summary: Increased expression of Kv1.3 in activated microglia is associated with neuroinflammation in Alzheimer's disease (AD), and inhibiting Kv1.3 has shown potential in improving cognitive function in AD mouse models. In this study, microglial expression of Kv1.3 was found to be increased in an AD animal model, and treatment with the Kv1.3 blocker HsTX1[R14A] significantly improved cognitive deficits. Transcriptomics analysis revealed changes in gene expression related to inflammation, neuron differentiation, synapse function, learning, and memory. These results highlight the potential of Kv1.3 blockade as a therapeutic strategy for AD.

Increased In Vivo Exposure of N-(4-Hydroxyphenyl) Retinamide (4-HPR) to Achieve Plasma Concentrations Effective against Dengue Virus

Summary: This study evaluated the in vivo exposure of 4-HPR and developed lipid-based formulations to enhance its exposure. Pharmacokinetic analysis showed that clinical formulation achieved effective plasma concentrations, while novel formulations achieved more than 3-fold improvement. However, solubility and first-pass intestinal elimination limited the exposure, which could be improved through CYP inhibition. Simulated exposure profiles suggested that b.i.d dosage regime with clinical formulation and new formulations/CYP inhibition were viable options to increase exposure.

A Homochiral Poly(2-oxazoline)-based Membrane for Efficient Enantioselective Separation

Summary: Chiral separation membranes have great potential for separating racemic mixtures into enantiopure components, but fabricating scalable membranes with both high selectivity and flux is challenging. In this study, a new type of enantioselective membrane was prepared using enantiopure S-poly(2,4-dimethyl-2-oxazoline) macromonomers and supported by graphene oxide nanosheets. The S-PdMeOx-based membrane showed near-quantitative enantiomeric excess and a high flux. This work demonstrates the potential of homochiral polymers in chiral discrimination and provides a new route to develop highly efficient enantioselective membranes.
