Identification of a Fibroblast-Related Prognostic Model in Glioma Based on Bioinformatics Methods

This study identified prognostic fibroblast-related genes in glioma and generated a novel risk signature that can independently predict the overall survival of glioma patients. Additionally, the study revealed important findings related to immune response, tumor microenvironment, and chemotherapy sensitivity in glioma.
Background: Glioma is the most common primary tumor of the central nervous system with a high lethality rate. This study aims to mine fibroblast-related genes with prognostic value and construct a corresponding prognostic model. Methods: A glioma-related TCGA (The Cancer Genome Atlas) cohort and a CGGA (Chinese Glioma Genome Atlas) cohort were incorporated into this study. Variance expression profiling was executed via the limma R package. The clusterProfiler R package was applied to perform a GO (Gene Ontology) analysis. The Kaplan-Meier (K-M) curve, LASSO regression analysis, and Cox analyses were implemented to determine the prognostic genes. A fibroblast-related risk model was created and affirmed by independent cohorts. We derived enriched pathways between the fibroblast-related high- and low-risk subgroups using gene set variation analysis (GSEA). The immune infiltration cell and the stromal cell were calculated using the microenvironment cell populations-counter (MCP-counter) method, and the immunotherapy response was assessed with the SubMap algorithm. The chemotherapy sensitivity was estimated using the pRRophetic R package. Results: A total of 93 differentially expressed fibroblast-related genes (DEFRGs) were uncovered in glioma. Seven prognostic genes were filtered out to create a fibroblast-related gene signature in the TCGA-glioma cohort training set. We then affirmed the fibroblast-related risk model via TCGA-glioma cohort and CGGA-glioma cohort testing sets. The Cox regression analysis proved that the fibroblast-related risk score was an independent prognostic predictor in prediction of the overall survival of glioma patients. The fibroblast-related gene signature revealed by the GSEA was applicable to the immune-relevant pathways. The MCP-counter algorithm results pointed to significant distinctions in the tumor microenvironment between fibroblast-related high- and low-risk subgroups. The SubMap analysis proved that the fibroblast-related risk score could predict the clinical sensitivity of immunotherapy. The chemotherapy sensitivity analysis indicated that low-risk patients were more sensitive to multiple chemotherapeutic drugs. Conclusion: Our study identified prognostic fibroblast-related genes and generated a novel risk signature that could evaluate the prognosis of glioma and offer a theoretical basis for clinical glioma therapy.

Summary: With the shift from the histology epoch to the molecular era in malignant glioma research, there is a growing need to investigate diagnostic/prognostic/therapeutic biomarkers and their regulatory mechanisms. This study identified differentially expressed genes and ceRNA networks in malignant glioma and constructed predictive models. The results revealed correlations between these genes and circadian rhythm, tumor immune microenvironment, and cellular senescence pathways. These findings provide new insights for precise diagnosis and personalized treatment of malignant glioma.

Summary: In this study, differentially expressed proteins (DEPs) and differentially expressed phosphoproteins (DEPPs) in the early and late stages of TBI were identified using TMT labeling, Phos affinity enrichment, and high-resolution LC-MS/MS analysis. Integrative analyses revealed the different and similar pathophysiologic mechanisms between the early and late stages, providing insights into useful biomarkers and underlying mechanisms in TBI and its sequelae.

Quantitative proteomic and phosphoproteomic analyses of the hippocampus reveal the involvement of NMDAR1 signaling in repetitive mild traumatic brain injury

Summary: Repetitive mild traumatic brain injury can lead to neurodegeneration and cognitive impairment, mainly due to damage to the hippocampus. This study found that mice exhibited cognitive impairment, increased p-tau expression, and decreased neuronal dendritic spines density in the hippocampus after repetitive mild traumatic brain injury. Proteomic and phosphoproteomic analyses revealed that inflammation, immunity, and coagulation were involved in the pathological changes in the hippocampus, while pathways related to neuronal function and structure were more consistent with neurodegeneration. N-methyl-D-aspartate receptor 1 was identified as a key molecule in the response to repetitive mild traumatic brain injury and its inhibition improved cognitive impairment. This suggests that N-methyl-D-aspartate receptor 1 signaling in the hippocampus plays a role in cognitive impairment in the chronic stage after repetitive mild traumatic brain injury and may serve as a potential target for intervention and treatment.

Role of Sirtuin 3 in Degenerative Diseases of the Central Nervous System

Summary: Sirt3, an NAD(+)-dependent deacetylase, plays an important role in the metabolic processes of mitochondria, including energy generation, the tricarboxylic acid cycle, and oxidative stress. Activation of Sirt3 has a strong neuroprotective impact by slowing down or preventing mitochondrial dysfunction in neurodegenerative disorders. The regulatory factors of Sirt3 in neurodegenerative diseases include antiapoptosis, oxidative stress, and the maintenance of metabolic homeostasis.

Incidence of Stress-Induced Hyperglycemia in Acute Ischemic Stroke: A Systematic Review and Meta-Analysis

Summary: The aim of this study was to evaluate the incidence of stress-induced hyperglycemia (SIH) in acute ischemic stroke (AIS). A total of 13 studies involving 4552 patients were included in the meta-analysis. The results showed that the incidence of SIH in AIS was 24%, with higher rates observed in North America compared to Europe and Asia. The study also found that the incidence of SIH decreased in studies published after 2010. These findings highlight the importance of prevention and control of SIH in AIS.
