Summary: E2F1 plays a significant role in maintaining gastric cancer stemness properties and is closely associated with clinical characteristics and prognosis in gastric cancer patients. High expression of E2F1 is correlated with worse overall survival, suggesting its potential as a prognostic biomarker and therapeutic target in gastric cancer.

Summary: The fate decisions of stem cells depend on signals from their microenvironment. By studying corneal epithelial stem cells, it was found that the unique biomechanical property of the niche supports the nuclear localization and function of Yes-associated protein (YAP). These findings suggest that stem cells can sense biomechanical niche signals, and manipulating the mechanosensory machinery could have implications for stem cell expansion in regenerative therapy.

Summary: The role and action mechanism of CDK5RAP2 in oral squamous cell carcinoma (OSCC) have not been fully understood. It is highly expressed in OSCC and its expression correlates with clinical stage and lymph node metastasis. CDK5RAP2 expression is regulated by the Wnt signaling pathway. Depletion of CDK5RAP2 inhibits OSCC tumorigenesis and migration, and disrupts spindle orientation during mitosis, indicating its potential as a CSC marker.

Summary: The upregulation of USP47 is associated with tumorigenesis and poor prognosis in clinical patients with colorectal cancer and is closely involved in the maintenance of the stemness of colorectal cancer stem cells (CCSCs). Silencing USP47 reduces proliferation and migration of colorectal cancer cells, suppresses the self-renewal of CCSCs, and induces apoptosis of CCSCs. Parthenolide (PTL) as a novel USP47 inhibitor shows potential in targeting CCSCs for colorectal cancer treatment.

Summary: The study demonstrates that PARK7 plays a crucial role in maintaining stemness and therapeutic resistance in GSCs. Knockdown of PARK7 inhibits self-renewal and invasion abilities of GSCs, increases sensitivity to ionizing radiation, and suppresses the expression of GSC signatures. Overexpression of PARK7 in non-GSCs enhances self-renewal activities, migration, and IR resistance.

Summary: This study identified JMJD6 as a novel modulator of TAM activation and demonstrated its involvement in the immunosuppressive activities of TAMs. JMJD6 deficiency reversed M2-like activation of macrophages, attenuated tumor growth, and increased tumor sensitivity to immune checkpoint blockades (ICBs). The inhibition of M2 polarization by JMJD6 was potentially mediated by the STAT3/IL-10 signaling pathway. These findings highlight the potential therapeutic role of targeting JMJD6/STAT3/IL-10 axis to enhance the effectiveness of ICBs in cancer treatment.

Summary: The study introduced an absolute REO-based stemness index to evaluate the stemness, showing higher correlation with culture time compared to previous methods. The stemness index demonstrated significant differences between cancer and normal tissues, as well as the ability to reveal intratumor heterogeneity at the stemness level. Higher stemness index was associated with poorer prognosis and greater oncogenic dedifferentiation, indicating its potential therapeutic and diagnostic implications.

Summary: In this study, the researchers collected and analyzed the expression data of RAD51AP1 in various cancer types. They found that RAD51AP1 was over-expressed in multiple cancer types and correlated with poor prognosis in certain cancers. They also demonstrated that RAD51AP1 promoted cancer cell proliferation and had broad associations with tumor stemness and the tumor immune microenvironment. These findings suggest that RAD51AP1 plays a significant role in carcinogenesis.

Summary: The novel lncRNA lnc408 functions as an oncogenic factor by inhibiting CBY1 expression and promoting beta-catenin accumulation in the nucleus, contributing to the maintenance of breast cancer stem cells (BCSCs) and tumorigenesis. This lnc408-CBY1-beta-catenin signaling axis may serve as a potential diagnostic and therapeutic target for breast cancer.

Summary: The study demonstrates that miR-200c-3p can promote proliferation and maintain stemness of ASCs, while also modulating the expression of immune regulatory proteins. These findings suggest the potential use of miR-200c-3p as a switch for ASCs reprogramming and clinical application.

Summary: Oral cancer is commonly associated with areca nut chewing, leading to the development of premalignant disease. Bacterial alterations in the oral microbiota may play a role in oral cancer progression. Studying microbial changes in patients with oral squamous cell carcinomas and oral submucous fibrosis (OSF) associated with areca nut chewing could provide valuable insights for the disease's pathobiology and potential diagnostic and treatment strategies.

Summary: The expression of CSNK1D was found to be elevated in GBM tissues compared with normal brain. CSNK1D high expression was associated with worse prognosis, enhanced stemness, and invasiveness in GSCs. Therefore, CSNK1D may serve as a potential therapeutic target for GBM patients.
