The bacteria express long-chain OAg in the intestinal lumen for increased resistance to antimicrobial peptides, but reduce OAg chain length upon sensing epithelial cell attachment, leading to enhanced injection of T3SS effectors into epithelial cells for host cell infection. This regulatory mechanism may be widespread in pathogenic bacteria and closely related to bacterial pathogenesis.

Summary: This study reveals that EHEC O157:H7 acquired a previously uncharacterized regulatory system rbfSR, which senses riboflavin produced by gut microbiota and activates the expression of virulence genes. This acquisition represents an essential step in the evolution of the highly pathogenic O157:H7. The introduction of rbfSR into other EHEC serotypes also increased their pathogenicity, indicating a potential public health issue.
