Photoswitchable polyurethane based nanoaggregates for on-command release of noncovalent guest molecules

This article reports the self-assembly behavior of light responsive amphiphilic polymers in aqueous milieu. The polymer forms stable micellar nanostructures and shows high encapsulation stability for hydrophobic guests. The release of encapsulated drugs can be controlled by UV light-induced changes in the hydrophobic environment and molecular arrangement of the micellar core. In addition, the polymer can modulate its surface charge in response to tumor-relevant pH, which has potential applications in targeted and controlled drug delivery systems.
The self-assembly of light responsive amphiphilic polymers has been of great interest in spatially and temporally controlled drug delivery applications. In this article, we report the design, synthesis and characterization of amphiphilic main chain azobenzene polyurethane as well as its self-assembly in aqueous milieu. This polymer self-assembles into micellar nanostructure (investigated by dynamic light scattering and scanning electron microscopy imaging) and shows hydrophobic guest sequestration with high encapsulation stability (probed by FRET experiment). The photoswitching (trans-to-cis) of these nanoaggregates has been investigated by irradiation with UV light (lambda = 365 nm), which results significant change in the hydrophobic environment and molecular arrangement in the micellar core. This leads to encapsulated guest release in a controlled manner as probed by UV-vis spectroscopy. Furthermore, we demonstrated tumor-relevant pH (similar to 6.5-6.8) induced surface charge modulation (neutral to positive) by zeta potential measurements. The light responsive guest release and pH-specific charge modulation, we believe, will have significant impact in development of delivery systems for targeted and controlled delivery of drug molecules.

Summary: The role of nonpolar co-solvents in initiating supramolecular polymerization and forming 1D nanowires has been studied by synthesizing a fluorophore-spacer-receptor derivative. The efficient gel formation in non-polar solvents is supported by microscopic and rheological data. Analysis of co-solvent composition reveals the crucial role of a critical co-solvent composition in initiating the polymerization process.

Tumor acidity-induced surface charge modulation in covalent nanonetworks for activated cellular uptake: targeted delivery of anticancer drugs and selective cancer cell death

Summary: A covalently cross-linked nanoassembly, called nanonetwork (NN), was designed and synthesized, which showed dual pH responsiveness for stable sequestration and sustained release of drug molecules in response to endosomal pH. The nanonetwork, fabricated from an amphiphile integrated with tertiary amine and acrylate functionalities, exhibited entropically driven self-assembly and micellar nanostructures. The cross-linking of the micellar core with thiol-acrylate Michael addition click reaction generated acid labile β-thioester functionalities, enabling slow hydrolysis and sustained release of the anti-cancer drug doxorubicin at endosomal pH.
