Molecular imaging of fibroblast activity in pressure overload heart failure using [68 Ga]Ga-FAPI-04 PET/CT

[(68) Ga]Ga-FAPI-04 PET/CT can monitor early cardiac fibrosis in pressure overload heart failure and predict the deterioration of cardiac function.
Purpose We aimed to evaluate whether [(68) Ga]Ga-FAPI-04 PET/CT could characterize the early stages of cardiac fibrosis in pressure overload heart failure. Methods Sprague-Dawley rats underwent abdominal aortic constriction (AAC) (n = 12) and sham surgery (n = 10). All rats were scanned with [(68) Ga]Ga-FAPI-04 PET/CT at 2, 4, and 8 weeks after surgery. The expression of fibroblast activation protein (FAP) in the myocardium was detected by immunohistochemistry. [(68) Ga]Ga-FAPI-04 PET signal and FAP expression were compared between two groups. Results Compared with the sham group, the AAC group presented with decreased ejection fraction (EF) and fractional shortening (FS) and increased left ventricular internal dimensions in diastole (LVIDd) and systole (LVIDs) at 4 and 8 weeks (all p < 0.01). The AAC group showed higher [(68) Ga]Ga-FAPI-04 accumulation in the heart than the sham group at 2, 4, and 8 weeks, and FAPI increased significantly from 2 to 8 weeks (all p < 0.001). Immunohistochemistry confirmed the higher density of the FAP(+) area in the AAC group. The intensity of the [(68) Ga]Ga-FAPI-04 correlated with the density of the FAP(+) area (p < 0.001). The expression of the [(68) Ga]Ga-FAPI-04 at 4 weeks correlated with the deterioration of cardiac function at 8 weeks (EF: R = - 0.87; FS: R = - 0.72; LVIDd: R = 0.77; LVIDs: R = 0.79; all p < 0.001). The AAC group also showed an increased [(68) Ga]Ga-FAPI-04 signal in the liver, peaking at 4 weeks and then declining. Cardiac and liver PET signals correlated at 4 weeks in the AAC group (R = 0.69, p = 0.0010), suggesting an early fibrotic link between organs. A combination of the [(68) Ga]Ga-FAPI-04 intensity in the heart and liver at 4 weeks better predicted the deterioration of cardiac function at 8 weeks. Conclusions The activated fibroblasts in the heart and liver after pressure overload can be monitored by [(68) Ga]Ga-FAPI-04 PET/CT, which reveals an early fibrotic link in cardio-liver interactions and could better predict nonischemic heart failure prognosis.

An Increase in Admission RDW Value Is Associated with Excess Short-Term Mortality Rates in Patients with Severe Burns

Summary: This study aimed to investigate the association between admission red blood cell distribution width (RDW) and outcomes in severely burned patients. The results showed that patients with high admission RDW value were more likely to die. Multivariate analysis revealed that higher admission RDW, age, full-thickness burned area, and inhalation injury were independent risk factors with 30-day mortality. A nomogram based on these risk factors was established to predict survival probability in severe burn patients.

Morbid obesity impacts mortality among inpatients with type a aortic dissection: an analysis of the national inpatient sample

The results showed that morbid obesity was associated with increased risk of in-hospital mortality and higher total costs in T(A)AD patients, and the prevalence of morbid obesity among T(A)AD inpatients has been increasing over the years.

Downregulation of NUP93 aggravates hypoxia-induced death of cardiomyocytes in vitro through abnormal regulation of gene transcription

Summary: The nuclear pore complex in the nuclear envelope plays a significant role in regulating the transportation of macromolecules between the nucleus and cytoplasm. This study investigated the impact of myocardial infarction on the expression and function of nucleoporins in cardiomyocytes. Knockdown of Nup93, Nup210, and Nup214 increased the expression of ANP and BNP, molecular markers of cardiomyocyte function. Knockdown of Nup93 aggravated hypoxia-induced cell injury and death, while overexpression of Nup93 had the opposite effect. This study reveals a new mechanism of Nup93 and provides potential therapeutic targets for ischemia-induced heart failure.

The HIF-1α/miR-26a-5p/PFKFB3/ULK1/2 axis regulates vascular remodeling in hypoxia-induced pulmonary hypertension by modulation of autophagy

Summary: This study found that miR-26a-5p is downregulated in the plasma of PAH patients, as well as in lung tissues and hypoxia-induced pulmonary arterial smooth muscle cells. HIF-1a specifically interacts with the promoter of miR-26a-5p and inhibits its expression in PASMCs. miR-26a-5p inhibits PASMC autophagy and proliferation by targeting PFKFB3, ULK1, and ULK2. Furthermore, overexpression of miR-26a-5p alleviates pulmonary vascular remodeling and right ventricular hypertrophy in hypoxia-induced PAH rat models.

State Residualisation and Kron Reduction for Model Order Reduction of Energy Systems

Summary: Due to the development of renewable energy sources, high-voltage transmission, and PE-interfaced loads, there is a growing number of power electronics converters being connected to energy systems. Modelling approximations commonly used for transient simulations of transmission systems may not be optimal for future grids, as power electronics-based devices and synchronous machines have different dynamic behaviours. To overcome the resource-demanding and time-consuming nature of full electromagnetic transient simulations, this paper proposes a combination of an event-based state residualisation approximation and the Kron reduction technique, which allows accurate transient simulations with a reduced number of state variables and simplified power systems equations for accelerated simulations.

Research on Fatigue Performance of Shape-Memory Alloy Bars under Low Cyclic Loading

Decreased expression of RPL15 and RPL18 exacerbated the calcification of valve interstitial cells during aortic valve calcification

Summary: This study aims to identify differentially expressed genes (DEGs) in calcified aortic valve tissues and analyze their correlation with clinical features in CAVD patients. A total of 1048 DEGs were identified, including 227 upregulated mRNAs and 821 downregulated mRNAs. RPL15 and RPL18 were found to be significantly decreased in calcified aortic valve tissues and negatively correlated with the osteogenic differentiation marker OPN in CAVD patients. Inhibition of RPL15 or RPL18 exacerbated valve interstitial cell calcification. These findings provide valuable clues for finding therapeutic targets for CAVD.

Summary: This study successfully implanted a chimney-shaped artificial mechanical heart valve, which fully accommodates the movement of the valve leaflets and allows for supra-annular surgical implantation. The artificial valve demonstrated normal mechanical properties and no obvious thromboembolism or abnormalities. It shows good wear resistance, histocompatibility, and antithrombotic and hemodynamic performance.
