In traditional Chinese medicine, the herbs that regulate blood play a vital role. Here, nine herbs including Typhae Pollen, Notoginseng Root, Common Bletilla Tuber, India Madder Root and Rhizome, Chinese Arborvitae Twig, Lignum Dalbergiae Oderiferae, Chuanxiong Rhizoma, Corydalis Tuber, and Motherwort Herb were selected and reviewed for their recent studies on anti-tumor, anti-inflammatory and cardiovascular effects. Besides, the analytical methods developed to qualify or quantify the active compounds of the herbs are also summarized. Copyright (C) 2015, Food and Drug Administration, Taiwan. Antiinflammatory triterpenoids from the fruiting bodies of Fomitopsis pinicola

Ping-Chung Kuo, Shih-Huang Tai, Ching-Che Hung, Tsong-Long Hwang, Liang-Mou Kuo, Sio Hong Lam, Kun-Ching Cheng, Daih-Huang Kuo, Hsin-Yi Hung, Tian-Shung Wu

Summary: A total of twelve undescribed lanostane-type triterpenes and twenty-two known triterpenes were isolated and identified from the medicinal bracket fungus Fomitopsis pinicola. Among the thirty-two triterpene compounds evaluated for antiinflammatory potential, pinicolasin J showed the highest potency as an inhibitor of superoxide anion generation and elastase release. This study contributes to the understanding of the nutritional and medicinal properties of Fomitopsis pinicola.

The study on structure-activity relationship between chromone derivatives and inhibition of superoxide anion generating from human neutrophils

Yi-Han Chang, Fang Shu-Yen, Hsuan-Yu Lai, Tsong-Long Hwang, Hsin-Yi Hung

Summary: The over activation of neutrophils has been linked to several inflammatory diseases, with the generation and release of superoxide anion being a critical pathologic mechanism. A compound, 2-(3,5-dimethoxyphenoxy)-5,7-dimethoxy-chromen-4-one, showed moderate inhibitory effect on superoxide anion generation, leading to the design and synthesis of chromones substituted with phenols or 3-flourothiophenol for suppression of superoxide anion generation. One successful example, 2-(3-fluorophenyl)sulfanyl-7-methoxy-chromen-4-one, exhibited significant anti-inflammatory effects with an IC50 value of 5.0 +/- 1.4 μM against superoxide anion generation.

Betulinic acid-nucleoside hybrid prevents acute alcohol -induced liver damage by promoting anti-oxidative stress and autophagy

Li-yun Zheng, Xi Zou, Yan-li Wang, Min Zou, Fang Ma, Ning Wang, Jia-wen Li, Ming-sheng Wang, Hsin-Yi Hung, Qiang Wang

Summary: Alcoholic abuse is a major cause of liver diseases globally, with oxidative stress being a critical factor. This study showed that BAN, a betulinic acid and nucleoside hybrid compound with good water solubility, effectively reversed acute alcohol-induced liver damages by reducing serum ALT level and liver oxidative stress. The protective role of BAN was mediated through hepatic FXR activation and the promotion of cellular autophagy.

Discovery of 5,7-Dimethoxy-2-(3,4,5-trimethoxyphenoxy)-chromen-4-one with Lipid Lowering Effects in Hepatocytes

Yi-Han Chang, Chia-Hung Yen, Chih-Chung Lai, Hsuan-Yu Lai, Hsin-Yi Hung

Summary: The population with nonalcoholic fatty liver disease (NAFLD) and nonalcoholic steatohepatitis (NASH) is increasing, and there is an urgent need to develop a new drug to overcome these diseases. Capillarisin and its related compounds were found to have protective effects against lipid droplet formation in liver cells. Compounds with trimethoxy groups on the phenyl ring showed better inhibitory properties, but some compounds exhibited cytotoxicity. Finally, an active compound was identified that reduced lipid droplet accumulation by up-regulating a specific protein, showing promise for managing NAFLD and NASH.

Exploration and biological evaluation of 7-methoxy-3-methyl-1H-chromeno [4,3-c]pyrazol-4-one as an activating transcription factor 3 inducer for managing metabolic syndrome

Yi-Han Chang, Heng Lin, Hsiao-Fen Li, Hsi-Hsien Chen, Hsin-Yi Hung

Summary: The induction of activating transcription factor 3 (ATF3) shows promise as a therapeutic mechanism to overcome metabolic syndrome. A study conducted a structure-activity relationship campaign and identified achiral 16c as a potential ATF3 inducer with lipid-lowering effects. In animal experiments, mice treated with 16c showed significant weight loss, reduced white adipose mass, and improved glycemic profile compared to the chiral lead compound 1b. Additionally, 16c ameliorated liver function and improved the plasma triglyceride profile. Therefore, 16c could be further developed as an ATF3 stimulant to alleviate metabolic syndrome.

Age-related differences in ERP correlates of value-based decision making

Poyu Chen, Hsin-Yi Hung, Joshua Oon Soo Goh

Summary: This study examines age-related differences in the temporal dynamics of neural processing in decision-making. Using a lottery-choice task and event-related potentials, the researchers found that younger adults showed greater P2 ERP-response positivity and later P3 positivity, which increased with win probability. Older adults displayed lower P2 responses and P3 amplitudes, with higher positivity for extreme win probabilities. Both age groups exhibited similar reward prediction error representations and salience integration. However, older adults showed more complex sensitivity to expectancy violations, indicating subjective uncertainty about reward expectations. The findings suggest that older adults' risk-taking behaviors may be influenced by reduced early neural processing of objective stimulus value and increased reliance on subjective processes.
